The carpel is a unique organ of angiosperms,a product of the evolution of the plant kingdom,and a major component of the ovary.The CLAVATA3(CLV3)gene is an important gene regulating the number of carpels(chambers)in plants,and is of great significance for horticultural plants where fruits are used for commercial organs and ornamental purposes.This study used bioinformatics analysis methods to search information,phylogenetic analysis and structural comparison of 11 horticultural plant(CLV3)genes,which laid a foundation for elucidating the evolutionary relationship between CLV3 genes in different species.Most studies suggest that plant carpels(compartments)are regulated by recessive genes.As a large-scale horticultural plant in China,cucumber has important edible value,and different fruit traits and color of fruit peel have great ornamental value.In this study,cucumber was selected as the material for constructing the5 carpel infiltration system.Multi-generation backcrossing was carried out by molecular marker-assisted selection method,and the Cs CLAVATA3 gene was genetically evaluated to further analyze the practicability of Cs CLAVATA3 gene to regulate the shape and weight of cucumber fruit.Its molecular mechanism lays the foundation.The main results of this study are as follows:1.According to the retrieved CLV3 genes of 11 species including Cucumis sativus,Arabidopsis thaliana,Solanum lycopersicum,Daucus carota,Malus domestica,Citrus sinensis,Fragaria vesca,Prunus persica,Mimulus guttatus,Aquilegia coerulea,and Capsella grandiflora,Signal peptide prediction,gene molecular weight,isoelectric point and other indicators were predicted,and the CLV3 gene was mapped on the chromosomes of each species.Of all the CLV3 genes retrieved,66 had signal peptides and 56 did not predict the presence of signal peptides.Predictive analysis of the physical and chemical properties of the CLV3 polypeptide gene family of each species revealed that the CLV3 gene has an amino acid size between72 and 229,with an average of the largest Malus domestica and the smallest Arabidopsis thaliana;the average molecular weight is the largest Malus domestica and the smallest is Fragaria vesca;The maximum protein fat coefficient is Mimulus guttatus,and the smallest is Fragaria vesca.2.The sequences of 11 species were compared and the phylogenetic tree was constructed by MEGA software.The analysis showed that the 122 genes from 11 species can be divided into two branches.The first group consists of 9 subgroups: including 40 pairs of homologous genes,of which 33 pairs belong to orthologous genes and 7 pairs belong to paralogous genes.To further analyze the relationship between the CLV3 genes of each species,a gene structure map was constructed in which all genes retrieved from Daucus carota,Malus domestica and Fragaria vesca contained only the CDS region.Among the many CLV3 genes,the CDS region of the MDP0000252090 gene in apples has the longest length and does not contain the UTR region.3.Taking cucumber as an example,genetic evaluation of the carpel number gene was carried out by constructing an infiltration system.By screening SSR molecular markers for polymorphism between parents,101 pairs of primers showed polymorphism between JC4 and TL(JT),and 95 pairs of primers showed polymorphism between CCMC and TL(CT).The d CAPs marker D1 of cucumber carpel number gene was used to select the progeny of different generations of the population,and the offspring containing the carpel number gene were selected.Using SSRs markers for background selection,in the BC1F1 backcross generation,the background recovery rate of each individual JT population ranged from61.9%-84.7%,the average genetic background recovery rate was 73.9%,and the background recovery rate of the five selected single plants was introduced.Between 78.2% and 84.7%,the average genetic background recovery rate was 80.9%,which was 5.9% higher than the theoretical value.In the BC2F1 backcross generation,the average genetic background recovery rate of selected individuals in the JT population was 96.4%,which was more than the theoretical value.8.9% higher,the average background recovery rate of selected plants in the CT population was 93.8%,which was 6.3% higher than the theoretical value.In the BC3F1 backcross generation,the genome-wide genetic background recovery rate of the two populations had reached 98%.The theoretical value is 4.3% higher,which lays a foundation for the infiltration system of the carpel number gene.
