Auricularia auricula-judge is one of the edible fungi with the largest cultivated area and consumption in China.At present,there are about 40 cultivars commonly cultivated.The quality of Auricularia auricula-judae is affected by both germplasm resources and ecological environment.JinJiang's Jelly ear,produced in Jinjiang Town,Shangri-la County,Yunnan Province,is a unique Auricularia auricula-judae species in Yunnan Province.Its edible taste and price are significantly higher than those of other Auricularia auricula-judae species.In this paper,ecological investigation,isolation and identification of germplasm resources and screening of cultivation formula were carried out.The main results of the thesis are as follows:The ecological environment of the collection site was investigated.After observing the morphology,basidiospore size and ITS molecular identification of Jelly ear,it was determined that it belonged to Auricularia auricula species.By Inoculating Different Carbon sources,nitrogen sources,inorganic salts,pH and other media,the media formula suitable for the growth of strain was screened out.The following are the results of media selection formula:Among the 10 carbon source screening media,the strain grew best in C6 medium.;Among the 7 nitrogen source screening media,the strain grew fastest and densest in N3 medium,and W1 medium was the most suitable for strain growth in 4 inorganic salt screening media.The strain growth on P4 medium was much better than that on other media formulas,and the strain growth on B3 medium was the best among the five media formulas.Rice bran and wheat bran were used as nitrogen sources.Corn cob,cottonseed hull,sawdust and sawdust are used as carbon source materials.Eight formulations,Y1,Y2,M1,M2,X1,X2,J1 and J2,were composed of them.By culturing in 8 kinds of formulas,the more suitable formula for the growth of strain was screened out.it was found that the growth of strain in the formula using wheat bran as nitrogen source was better than that in the formula using rice bran as nitrogen source.Among the four formulations with wheat bran as nitrogen source,the strain of J1 formulation grew fast,but the mycelium was sparse,and it was difficult to produce mushrooms in later stage,and the yield was very low.Y1,Ml and X1 formula mycelium grew slowly at the beginning,but increased significantly at the later stage,and the mycelium was white and dense.From the later stage of mushroom production,M1 formula is superior to other formulations,so Ml formula is the most suitable for mycelium growth.The situation of mushroom production is not ideal.The mushroom bags only produce one mushroom.Air humidity and temperature have great influence on mushroom production.The amount of mushroom production increases significantly in rainy season.After the temperature drops in October,the amount of mushroom production decreases sharply.Although the results of artificial cultivation are still unsatisfactory,the domesticated cultivation of JinJiang's Jelly ear has made a bold attempt to provide germplasm materials and technical exploration for subsequent large-scale artificial cultivation and industrial development.
