Picea mongolica is a rare and endangered spruce species in China.The natural population is only concentrated in the limited area of the east edge of Hunshandake sandy land and formed a sandy land forest ecosystem which is rare at home and abroad.P.mongolica played a very important role in the regional ecological protection system.However,the systematic status of the species has been controversial for many years.In order to clarify the phylogenetic status of P.mongolica,provide the basis of DNA level for the spruce classification,and provide theoretical and technical basis for the effective conservation and utilization of this spruce resource,the genetic diversity of P.mongolica was studied based on cpDNA and mtDNA sequence information,according to natural samples of P.mongolica and its nearby species Picea koraiensis,Picea meyeri and Picea wilsonii.The main results are as follows:?1?In the three spruce mitochondrial gene sequence,only one mitochondrial gene sequence has variation,four variation sites and four haplotypes were observed.The variation level of the mitochondrial gene sequence was relatively low,which may not be suitable for species delimitation.?2?The three chloroplast gene sequences of spruce were all have variation,and the polymorphism was high.A total of 10 mutation sites and 15 haplotypes were found.The spruce chloroplast gene sequence may be more suitable for the study of the species definition of the genus spruce because of the high polymorphism.?3?P.mongolica has the highest genetic diversity in four spruce species.The primary distribution area of P.mongolica Baiyinaobao and Baiyinxile may be the potential spruce divergence center.?4?According to the haplotype distribution of joint cpDNA,haplotypes may have evolved from Hap14 and Hap 15 to Hap9 and Hap13 then to Hap12.?5?According to the genetic diversity and haplotype of the chloroplast gene sequence,the genetic difference between the P.mongolica and its nearby species is significant,and the P.mongolica should be a taxonomical species.
