The multi-leaf type and standard type of clover are the F1 generation lines obtained by the hybridization and embryo rescue techniques of Mengnong clover No.1 as the female parent and white clover as the male parent.Since 2005,F1 generation plants that can be cultivated in greenhouses and fields have been obtained through a series of studies on the in vitro culture of Fi generation,the organogenesis and embryo germination of stem segments in vitro.During the research of hybrid breeding,some clover plants showed multi-leaf traits of 4-5 leaflet.Through the nutrient expansion of the roots of these plants,a series of multi-leaf strain are formed.This finding is of great significance for the cultivation of new varieties.In order to explore the molecular mechanism of the multi-leaf phenomenon of clover,one of the multi-leaf clover strains(1-1)and the standard clover strains(2-2)was selected as the experimental group and the control group,respectively.The group established three biological replicates.T1,T2 and T3 were samples of the test group,and T4,T5 and T6 were samples of the control group.Transcriptome sequencing analysis was performed on the 6 samples based on ma-seq technology.The main findings are as follows:(1)Transcriptome data analysis showed 1985 differentially expressed genes from multi-leaf clover and standard clover,including 794 up-regulated genes and 1191 down-regulated genes.The GO functional annotation indicates that differentially expressed genes are mainly enriched in processes such as protein phosphorylation,defense response,nucleus,cytosol,oxidoreductase activity and protein binding.(2)Among the differentially expressed genes,68 specific expression genes of the multi-leaf clover were found,and 121 specific expression genes of the standard type clover were found.KEGG enrichment analysis indicated that a specific expression gene(c68409.graph_c0)of the multi-leaf clover was enriched in homologous recombination,mismatch repair,DNA replication,and nucleotide excision repair signaling pathway.It suggests that this gene may be involved in biological processes such as DNA replication,nucleotide excision repair,homologous recombination and mismatch repair during cell meiosis.This provides a theoretical basis for further exploring the molecular mechanism of multi-leaf traits in the hybrids of Mengnong clover 1 and white clover.It is of great significance for the future deep excavation of potential genes affecting the stable inheritance of clover multi-leaf traits.
