Manglietia glanca Blume,is an evergreen broad-leaved tall tree belonging to the genus Manglietia of Magnoliaceae of Magnolia family.It is an excellent ornamental tree species in gardens.It also plays an important role in vegetation restoration and water conservation.However,introduced and cultivated in China,it is often abnormal species in seed setting,not full,difficult to meet the requirements of seedling.In asexual propagation,grafting,cutting,tissue culture are still in the experimental stage,the rooting rate is very low,can not meet the needs of large-scale factory seedling.Therefore,the factors affecting the survival of cutting are compared and optimized in this paper,and the relationship between physiological and biochemical changes of cuttings and rooting under the action of matrix,type of cuttings and rooting agent is discussed,and the relationship between rooting and physiological and biochemical changes of cuttings is discussed.In order to improve the level of cutting seedling of Manglietia glanca Blume.The findings are as follows:(1)Effect of cutting medium on cutting type of Manglietia glanca BlumeIn cutting medium selection,substrate V(coconut bran):V(pearlite):V(peat):V(yellow soil)=3:3:3:1 is the most suitable for cutting propagation of Manglietia glanca Blume,the highest rooting rate can reach 46.33%.The best index of root efficiency is 2.44.The rooting rate of the river sand in the control group is only 9%,and the root effect index is only 0.19.The different matrix formulations have an effect on the enzyme activity,the endogenous hormone and the nutrient content in the insertion process of the cuttings.The content of MDA,SOD and POD are low,and the activity of SOD and POD is high,which is beneficial to the rooting of the cuttings.In the endogenous hormone,the content of IAA,GA3,ZR and ABA are low,and the ratio of IAA/ABA is high,which is beneficial to the rooting of the cuttings.In nutrient content,soluble sugar and soluble protein content are high,which is beneficial to rooting of cuttings.(2)Effect of cuttings on cuttings of Manglietia glanca BlumeThe average rooting rate is 53.33%and the root efficiency index is 28.59.The rooting characteristics of cuttings with top buds are the best,and the rooting rate is 53.33%and the root efficiency index is 2.86.Among the four types of cuttings with top bud,upper branch cuttings,middle branch cuttings and lower branch cuttings,the lowest rooting rate and the lowest root effect index are 26%and 1 respectively.Different types of cuttings have the same trend of change in the process of cutting,but the amount of change is different,which has an effect on the rooting effect of cuttings.The initial value of MDA content in cuttings with apical bud is high,but the range of increase in rooting process is the smallest compared with the other three types of cuttings,which also makes the rooting effect of cuttings with apical buds better.At the same time,compared with the other three types of cuttings,the ratio of SOD,POD,IAA,IAA/ABA,soluble sugar and soluble protein,and the content of GA3,ZR and ABA in the cuttings with top bud are higher than those of the other three types of cuttings,which is beneficial to the rooting of cuttings.(3)The effect of rooting agent on the insertion of gray-wood lotusIn the orthogonal experiment,the highest rooting rate is 53.33%,the average adventitious root number is 6.33cm,the average root length is 4.97cm,the average root tip number is 107.3,and the rooting rate is 53.33%,the average root length is 4.97cm,and the average root tip number is 107.3 when the cuttings are soaked in 1000mg/L for 2h.The highest root index is 3.20.In the control group,the lowest rooting rate is 9.33%,the average adventitious root number is 1.87,the average root length is 1.70 cm,the average root tip is at least 27,and the root effect index is only 0.17.Hormone treatment can effectively promote rooting of cuttings.According to the range analysis,the order of the ability of the rooting rate of the cuttings to influence the rooting rate of the cuttings is as follows:the species of the rooting agent>the concentration of the rooting agent>the treatment time of the rooting agent,and the rooting effect is the best in the species of the rooting agent.The activity of SOD,the content of MDA and the activity of POD in the base of cuttings are closely related to rooting.After different rooting agents,the enzyme activities in cuttings varied with each other in the process of rooting.Compared with different treatments,the content of MDA in cuttings of Manglietia glanca Blume is lower,and the activity of SOD and POD is higher,which is beneficial to root growth and could improve the root phenotype characters of adventitious root length,indefinite number and root tip number of cuttings.The content of endogenous hormones in the base of cuttings is also related to rooting.The content of IAA is high,the ratio of IAA/ABA is high,the contents of GA3,ZR and ABA are low,which promoted the rooting of cuttings.Nutrients provide energy for rooting of cuttings,soluble sugar and soluble protein contents are at a higher level,and promote rooting of cuttings.
