Spodoptera litura,belonging to Noctuidae family in Lepidoptera order,is an important cruciferous vegetable pest and poses a serious threat to vegetable production in China.High-efficiency cyhalothrin has high efficiency and low toxicity,which is widely used in actual production.Due to long-term dependence and irregular use of chemicals for control,S.litura has developed a high level of resistance to most insecticides.In this paper,the RNA-seq,bioinformatics analysis and qRT-PCR analysis of S.litura were carried out on Illmina platform.The expression levels of multifunctional oxidase(P450s)gene and glutathione S-transferase(GSTs)gene of S.litura were studied under high-efficiency cypermethrin treatment,which provided the basis for further research on the mechanism of the resistance of S.litura to high-efficiency cyfluthrin.The following results have been obtained.1、RNA-seq of S.litura and Bioinformatics AnalysisThrough the RNA-seq of S.litura and the assembly of the obtained reads,38175 Unigene were obtained.The high quality data(Q20)of all samples were higher than 98.37%,the base ratio of Q30 was not lower than 95.57%,and the N50 was 1538bp.By analyzing the transcriptome data,67 P450 genes were screened and identified.These genes were named CYP4AU1-CYP428A1,followed by 8 GST genes and 8 esterase genes.2、Expression Level of P450 Gene and GST Gene in S.lituraThe mRNA levels of 17 P450 genes and 5 GST genes treated with different concentrations of cyhalothrin were detected by qRT-PCR.The results showed that the mRNA levels of P450 and GST genes of most S.litura were significantly different in different concentrations of cyhalothrin.Among P450 genes,2 genes showed a downward trend and 4 genes an upward and then downward trend.Among GST genes,2 genes displayed a downward trend.Through transcriptome information analysis and detoxification enzyme related research,this study provides a theoretical basis for detoxification enzyme in the development process of S.litura drug resistance and some new ideas for the prevention and control of S.litura.
