Infectious bovine rhinotracheitis virus(IBRV)is a serious pathogen of cattle,IBRV infection can induce Infectious bovine rhinotracheitis(IBR),and cause significant economic losses to the cattle industry,vaccines can greatly reduce the incidence of IBR and is the most cost-effective public health solution.To increase cell density and improve virus titer,Bioreactors are often used in vaccine industry.However most domestic companies still culture cell in low-efficiency rotary bottle way,thus the vaccine production and quality lagged behind the world's advanced level,which greatly limited the development of vaccine industry.In recent years,with the development of molecular biology,scientists began to explore the mechanism of interaction between viruses and host cells to control the processes of viral infection and replication,and finally increase virus titers.It is well known that cold atmospheric plasma play great function for strain mutagenesis and cancer treatment,but its enormous potential application remains to be discovered.In order to improve the yield of IBRV in MDBK cells,this study explored the effects of plasma treatment on IBRV production.The main research results were shown below:(1)A method was proposed for IBRV rapid detection and quantification.There is no significant difference among the detection results in boiling time of 10-30 minutes and the reproducibility of this method is better than using the column purification kit;(2)The effects of different plasma treatment time on IBRV yield was determined and the optimal time for plasma treatment was 2 min.We also compared the effects of plasma treatment individually for cells,viruses,and both.Finally we determined the best plasma treatment way was to treat cells and viruses simultaneously;(3)The reasons behind the different phenomena caused by different plasma treatment methods was explored,and that plasma treatment was proved to improve the infection ability of IBRV and promote the process of IBRV entering MDBK cells.We also confirmed that plasma treatment could inhibit the phagocytosis of MDBK cells and promote the expression of IFN-? in MDBK cells.ROS is only part of its functional composition and cannot completely replace the effect of plasma;moreover plasma treatment has certain specificity on the promotion of viral replication;(4)To study the inhibitory effect of BoIFN-? on IBRV replication,we successfully expressed BoIFN-? by using the C.glutamicum expression system,this recombinant protein has a good inhibitory effect on the proliferation of IBRV in MDBK cells.
