| As a carrier of important blood parasites,ticks often carry a large number of different pathogens.The disease spread by ticks is widespread throughout the world,seriously threat the health of animals and people.In order to further understand the infection status of blood parasites in some areas of Henan Province(Zhengzhou,Dengfeng and Luoyang),the epidemic characteristics of infection were mastered.The experiment was completed from March 2018 to the December.A total of 1441 blood samples were collected.This experiment used microscopic examination and conventional PCR methods to perform molecular biology of B.gibsoni(18S r RNA),H.canis(18S rRNA)and AP(16S rRNA)in dog blood samples from parts of Henan Province.The factors influencing were regional,gender,age,presence or absence of parasitic infections and seasons.1.Clinical characteristics and molecular genetics of B.gibsoniA total of 56 positive samples were detected which include 5 positive samples in Zhengzhou,51 positive samples in Dengfeng by microscopy in 1441 samples.And 46 of the canine blood samples showed significant clinical symptoms which include 31 samples of body temperature above 39 ℃(67.39%),15 samples of anemia or pale mucous mcmbranes(32.6%),9 samples of hemoglobinuria(19.56%).Based on 18S r RNA gene locus,a total of 162 positive samples were detected by PCR,and the infection rate is 11.24%.46 positive samples showing clinical symptoms were Babesia gigas,with a positive rate of 100%.The infection rate of Dengfeng was 43.44%,which is higher than that of Zhengzhou and Luoyang;autumn is the high incidence of this disease,the infection rate is 42.42%;the incidence of male samples was higher than that of females,which were 24.47%and 9.96%.The pets in Dengfeng have a higher probability of contact with arthropods such as ticks,and the chances of suffering from blood protozoa are high.2.Molecular genetics of Anaplasma phagocytophilumBased on the 16S r RNA locus,a total of 66 positive samples were detected by PCR and The infection AP rate was 4.58%(66/1441).The sample infection rate in Dengfeng is the highest,and the infection rate is 5.25%.The infection rate in autumn is 4.71%.The infection rates of male and female samples were 4.43%and 4.21%.In terms of regional and seasonal factors,AP and B.gibsoni have the same prevalence,and the infection rate is the highest in Dengfeng and autumn.In the microscopic examination,no pathogen of AP was found.3.molecular genetics of H.canis.The route of transmission of H.eanis is different from that of B.gibsoni and AP.It requires the host to ingest the arthropods such as ticks into the body and enter the host through the gastrointestinal tract.Based on the 18S r RNA geen locus,and only H.canis was detected in the samples from Dengfeng,with an infection rate of 4.08%.And there have no infection in spring,summer and winter,concentrated in autumn,the infection rate was 4.71%.The infection rates of male and female samples were 2.11%and 1.53%.The infection rate of H.canis is significantly lower than that of B.gibsoni,which has a greater relationship with the transmission pathway of H.canis.ConclusionThis text uses PCR detection methods to analyze the molecular epidemiology of B.gibsoni,AP and H.canis in Henan Province,and the evaluation of the molecular phylogenetic development of three blood parasitic.Therefore,the development of preventive measures has important significance guiding for preventing the spread of pathogens. |
