MicroRNAs(miRNAs)are small RNA regulatory factors that are widely present in eukaryltic cells and typically have a length of about 21 bases.It is cut from the stem loop precursor(pre-miRNA)and functions by completely matching or nearly completely matching the target gene and causing mRNA degradation or inhibiting translation,so they play an important role in plant growth and development as well as environmental stress.However,there is no research on environmental stress responsive miRNA in Ginkgo biloba L.This research focuses on the miRNAs in Ginkgo biloba L.to find the relevant miRNA in drought,ultraviolet,hormone stress by the method of bioinformatics analysis and high-throughput sequencing technology,analyze the differentlly expressed miRNA,screen known miRNA and forecast novel miRNA,deepening the understanding of the molecular mechanism of resistance in Ginkgo biloba L..We discuss the role of miRNA regulation in resistance of Ginkgo biloba L..The main research results can be summarized as follow:(1)CLG,UVG,DRG,CCG,CVG,CRG were sequenced using the high-throughput sequencing technology,which generated 7648427,7745539,7469773,11614517,6463373,7903059 reads respectively after data filtration.Reference sequence alignment analysis showed that the numbers of clean reads sequences of CCG,DRG,UVG were descend than CLG,CRG,CVG,which from 82.83% to 79.89%,from 81.37% to 78.46%,from 83.45% to 72.05%,respectively.This indicated that the miRNA molecule may play an important role in the endogenous regulation under chlormequat chloride,drought and ultraviolet stress.Known miRNA analysis showed that 12 miRNA mature bodies can be mapped,14 miRNA hairpins can be mapped,the species of sRNA mapped to the known miRNA are 412,and the number is 72829.(2)CCGvsCRG,miRNA differential expression analysis showed that a total of sRNA is 66,17 significant differential expressions,9 up-regulations,8 down-regulations.The result showed miRNA may play a role in Ginkgo biloba L.under CCC stress.DRGvsCRG,19 significant differential expressions,7 up-regulations,12 down down-regulations.The result showed the miRNA may play a role in Ginkgo biloba L.under drought stress.UVGvsCVG,34 significant differential expressions,11 up-regulations,23 down-regulations.The result showed the miRNA may play a role in Ginkgo biloba L.under ultraviolet stress.(3)Novel miRNA identification results show that 54 miRNA mature bodies can be predicted,41 miRNA* can be predicted,55 miRNA hairpins can be predicted,the species of sRNA mapped to the novel miRNA are 1898,and the number is 211058.(4)miRNA target genes functional annotation and GO analysis,the main functions of the target genes include translation,reproduction,signal transduction,transport of amino acids and carbohydrates and metabolism and so on.It proves that miRNA plays an important role in the growth and development of organisms,cell differentiation and metabolism.
