Growth hormone receptor(GHR),uncoupling protein family gene(UCPs),F-box protein 32(FBXO32)and Wingless-type MMTV integration site family(Wnt8a)play an important role in maintaining the balance and growth of normal physiological and biochemical mechanisms in animals.In order to verify whether 8 SNPs of 5 genes affect the growth traits of Angus cattle in Ningxia region.In this study,the SNP loci of GHR,UCP2,UCP3,FBX032 and Wnt8a genes were genotyped in 1126 Angus cattle aged 20 months approximately from 5 cattle farms in Ningxia.The genotyping of the SNP loci of GHR,UCP2,UCP3S FBXO32 and Wnt8a genes was carried out by Sequenom typing technique.The genetic information of the population related to each SNP locus was statistically analyzed,and the correlation analysis was carried out between SNPs and growth traits of Angus cattle.The results of the study are as follows:1.In this study,Sequenom typing technique was used to detect and analyze 8 SNPs genes,and we found the selected gene loci were polymorphic in the Angus cattle population in Ningxia.GHR-149,UCP2-655 and FBX032-53423 loci were all moderately polymorphic(0.25<PIC<0.5).All the mutation sites were found to be in Hardy-Weinberg equilibrium(P>0.05)after chi-square test except UCP3-5465 locus.2.There was a significant correlation between GHR-149(g.149A>G)locus and body height and cross height of Angus cattle.The mean body height and waist height of AA individuals were significantly higher than those of AG and GG type individuals(P<0.05).The UCP2-655(g.655G>A)locus was significantly correlated with the chest circximference and abdominal circumference of Angus cattle.UCP3-5465(g.5465A>G)locus was significantly correlated with the weight of Angus cattle(P<0.05),and the average chest circumference and abdominal circumference of AG type individuals were significantly higher than those of AA and GG type individuals(P<0.05).The mean body weight of G type individuals was significantly higher than that of AG type individuals(P<0.05),and the FBXO32-53423(g.53423A>G)locus was significantly correlated with the body oblique length of Angus cattle.The mean values of body oblique length of AA and AG individuals were significantly higher than those of GG type individuals(P<0.05).The Wnt8a-910(g.910A>G)locus had a significant effect on the chest circumference of Angus cattle,and the mean of AA genotype individuals was significantly higher than that of AG genotype individuals(P<0.05).3.Linkage disequilibrium and haplotype analysis showed that two loci of Wnt8a-244 and Wnt8a-910 were in complete linkage disequilibrlum,and three loci of UCP2-655,UCP3-7921 and UCP3-8018 were in strong linkage disequilibrium.The haplotype frequency of Hl(CA)in Blockl was the highest(96.8%).The haplotype frequency of H3(AGC)in Block2 was the highest(79.9%).The haplotype H3(AGC)composed of UCP2-655,UCP3-7921 and UCP3-8018 loci had significant effects on body height and chest circumference of Angus cattle(P<0.05).The results of this study are of great significance for further understanding and mastering the growth and development of Angus cattle after the introduction of Ningxia region,opening up a correct way of utilization,and selecting excellent individuals to play a better role in breed effect.It provides a reference for molecular marker assisted selection of Angus cattle in Ningxia.
