China's mulberry germplasm resources are abundant,and with the continuous collection and preservation,germplasm resources have been established in various regions.Resources provide a wide range of breeding materials.In order to better develop and utilize germplasm resources,the measurement and evaluation of mulberry germplasm resources is an indispensable link.In this paper,the pluripotency of Guangxi mulberry germplasm resources was determined,and the ploidy of the preserved germplasm resources was clarified.At the same time,the ploidy induction of mulberry seeds and mulberry seedlings was carried out by chemical methods,and the polyploid plants were selected from them.The growth of mulberry seedlings was observed after induction,and the law of ploidy induction of mulberry seedlings was preliminarily understood.Provide a certain theoretical basis for multiple mulberry treesThis paper mainly uses flow cytometry to determine mulberry ploidy.In the study,different leaf positions of mulberry trees,different parts of the same young leaves,leaf buds in the depilation stage,and radicles after seed germination were selected for flow cytometry.It was found that different materials were affected by flow cytometry.The experimental results showed that the ploidy effect was detected by flow cytometry:young leaves>first leaf leaf>second leaf leaf;young leaf leaf base>young leaf tail,leaf bud is an ideal ploidy detection material,but leaf bud sampling is not easy.According to the comprehensive factors,it is determined that the young leaves are selected as the mulberry ploidy detection materials.The main findings are as follows:Among the 527 mulberry germplasm resources tested,268 were diploid,154 were triploid,88 were tetraploid,and 17 were mixed.There were 9 diploid cells mixed with diploid cells and triploid cells,and 8 mixed with diploid cells and tetraploid cells.Five diploid,triploid and tetraploid mulberry trees were selected from the mulberry germplasm resources.The chlorophyll SPAD(Soil and Plant Analyzer Development,SPAD)value,soluble sugar content,and total protein concentration were measured at different time periods.The experimental results show that the chlorophyll SPAD value and total protein concentration and the regularity of the month are in late August<late September<late October<late November,The basic trend of soluble sugar content is from late September to late October ?late November<late August.The ploidy of F,plants of two different diploid mulberry trees and tetraploid mulberry hybrids was detected,and 100 strains were detected for each hybrid combination.The results showed that the F1 plants of the mulberry hybrid combination A accounted for 96%of the triploids,the diploids accounted for 4%,the F1 plants of the mulberry hybrid combination B accounted for 82%of the triploids,and the diploids accounted for 18%.Guisangyou 62(diploid)and Guisang 6(triploid)were induced by seed soaking method and seedling dropping method.The induced plants were subjected to ploidy determination by flow cytometry.The experimental results showed that the mutagenic rate of Guisangyou 62 induced by seed soaking method was 3.3%,and the mutagenic rate of Guisang 6 was 2.7%.The mutagenic rate of Guisangyou 62 induced by seedling dropping method was 11.3%,and the mutagenic rate of Guisang 6 was 8.2%.During the experiment,the growth of seedlings induced by seed soaking and the anatomy of seedlings were observed.It was found that the seedlings showed two different states during the early growth process.One was that the hypocotyls did not extend out of the ground to start plant growth,and the other was that the hypocotyls first extended out of the ground before plant growth.
