| circRNAs are a class of reverse-variable cleavage of mRNA precursors that are involved in the regulation of multiple life processes through cell type-specific expression and tissue-specific expression.Carya cathayensis Sarg male and female flower development exist great differences in time and space.In order to explore the expression pattern of circRNA during the differentiation of male and female flower buds of hickory,this study constructed a total of six sample circRNA-specific libraries in three stages of male and female flower buds development of hickory,using strand specific PE150 bp high-throughput.The differential circRNA target miRNA interaction was predicted.The expression of miRNA in female flower development was studied by qRT-PCR.Finally,the regulatory network map of circRNA flowering was constructed.The main results are as follows:(1)Sequencing of the three stages of female flower buds a total of 76.54 G filtered test data were obtained,and 4857 circRNAs were newly identified with reference to the hickory locus.The expression level was normalized by CPM and the source gene was annotated,and a total of 76 flower-related circRNAs were obtained in 4449 differentially expressed circRNAs.(2)The samples of the male flower buds in three periods were total obtained 76.3G clean data.Using the above method,a total of 5,517 circRNAs were identified.After normalizing the expression profile by CPM,there were 2209 differentially expressed circRNAs in the whole male flower bud differentiation process,of which 34 annotations were associated with flowering.(3)During flower bud formation,we predicted 10 different miRNAs in the female flower bud samples,including temperature-responsive miR164 and miR399 members,and231 differential circRNAs predicted to participate in the regulation of FT-interacting protein genes by miR167 sponge.Two target miRNAs(cca-miR408-3p、cca-miR2111a)were predicted to interact with differentially expressed circRNAs in male flower buds.(4)Quantitative experimental results of female flower bud flower developmentdifferentially expressed circRNA showed that most detection circRNA(8/12)coincided with the predicted trend of differential expression analysis.It indicates that circRNAs are involved in the regulation of female flower bud development. |
