Unfavorable environmental conditions such as oxidative stress,hypoxia and high temperature often cause the corresponding stress and the stress response of the organism,thus coping with toxic damage suffered by the body in the process to maintain the relative stability of the body's metabolism.The heat stress response of organisms caused by high temperature stress has adverse effects on the reproductive development and metabolism of animals.Dandelion is a traditional Chinese herbal medicine,and its various active substances make it have many aspects.Medicinal efficacy,this paper uses C.elegans as a model organism,using three different methods to extract total flavonoids from dandelion,qualitative and quantitative analysis to establish the best extraction method,and studied its lifespan and oxidation in the heat stress state of nematodes The influence of the stimulating ability to initially determine its role in heat resistance.The research results are as follows:1.Comparison of different extraction methods of total flavonoids from dandelionThe concentrations of total flavonoids in dandelion obtained by alcohol extraction,water extraction and alcohol extraction were 1.042 mg/mL,2.058 mg/mL and 2.884 mg/mL,respectively.The concentration of the extract obtained by the microwave method was the highest.However,after the obtained flavonoids were subjected to UV color reaction,aluminum trichloride reaction,magnesium acetate reaction and paper chromatography,it was found that the relative purity of the total flavonoids obtained by the water extraction and alcohol precipitation method was relatively high.2.Effect of total flavonoids of dandelion on the survival rate of heat stress nematodesThe total flavonoids of dandelion can improve the survival rate of nematodes under heat stress and exhibit a certain dose effect.The lifespan of nematodes in the0.625 mg/mL total flavonoid treatment group was significantly different from that in the blank group at 3 h and 4 h after heat stress,and the difference was significant at 2h heat stress treatment(P<0.01).3.Effects of total flavonoids from dandelion on the exercise ability of heat stress nematodesThe total flavonoids of dandelion have a certain enhancement effect on the head swinging ability and basic exercise ability of heat stress nematodes,and exhibit a certain dose effect.The difference of nematodes in 0.625 mg/mL total flavonoids treatment group was extremely significant(P<0.01);The sway frequency of nematodes in 1.25 mg/mL total flavonoid treatment group was significantly different from that of the control group,but compared with the other two experimental groups.The frequency under the duration is reduced.It is speculated that the total flavonoid concentration may cause a certain degree of damage to the worm.4.Effects of total flavonoids of dandelion on oxidative stress of heat stress nematodesCompared with the control group,the related enzyme activities in the dandelion flavonoids treatment group were improved,and the MDA content was decreased,indicating that the flavonoids of dandelion had a certain protective effect on the oxidative damage of nematodes,and showed a certain extent within a certain range.The dose effect;the heat stress treatment at 35 °C for 3 h,the lowest MDA content in the nematode and the highest SOD activity in the drug concentration of 0.625 mg/mL;0.3125 mg/mL,CAT in the nematode after 3 h of heat treatment at 35 °C,the enzyme activity is the highest.It is suggested that the protective effect of total flavonoids of dandelion on nematodes under heat stress may be related to the change of activity of related antioxidant enzymes.5.Effects of total flavonoids of dandelion on the expression of heat stress-related nematode related genesThe nematode cultured at 35 °C,3 h after heat stress,RNA was extracted and reverse transcribed.The expression of jnk-1 gene in the flavonoid treatment group was increased.The expression of 0.625 mg/mL was higher than that of the control group.The up-regulation was about 4.6 times;the expression of eat-2 gene was up-regulated by 13.14 times;when the expression level of gei-7 gene was 0.3125mg/mL,the comparison group was up-regulated by about 1.25 times,and the difference was significant(P<0.05).
