Mapping And Cloning Of The Dwarfing Gene Rht-dp In Dwarfing Polish Wheat

Dwarf Polish wheat?Triticum polonicum L.,DPW?from Turpan,Xinjiang,carries a recessive dwarfing gene Rht-dp on the 4BS chromosome,which is a GA-insensitive gene.In this study,the genetic analysis of Rht-dp was performed by using SLAF-Seq-BSA and SSR markers,with cloning and analysis in 56 tetraploid wheat.The main results are shown below:1.Statistical analysis of plant height of the DPW×HPW F₂?850 indivduals?:Rht-dp is a single gene.2.SLAF-Seq analysis of DPW×HPW F₇ RILs,Rht-dp was mapped on the 4B chromosome started from 154,766,751 to 343,858,300.3.13 pairs of polymorphic SSR markers were screened in the initial location,and linkage analysis revealed that 12 individuals did not linked to all markers.Further analysis and screening showed that 3 pairs of SSR polymorphism markers linked to these 12individuals.Linkage analysis was performed on DPW/HPW RILs F₇ and F₈,DPW/Aiganfanmai RILs F₆ population,and Rht-dp was mapped between Xgpw2994.1 and Xgpw3128.1.Taking the Chinese spring genome?v1.0-assembly?as the reference genome,Rht-dp is located in the 4BS chromosome 29,914,199-31,286,578.There are 5 coding genes in this region,and Rht-B1 encoding Rht1_wheat DELLA protein was used as a candidate gene.4.By cloning Rht-B1 in DPW and HPW,we found that the sequence of Rht-dp was consistent with the Rht-B1b.Based on the the difference between DPW and HPW sequences,we developed Rht-B1 Indel and made the linkage analysis in DPW/HPW RILs F₇ and F₈,DPW/Aiganfanmai RILs F₆.This marker was cosegregated with Rht-dp,which proved that the Rht-dp was Rht-B1b.Meanwhile,we tested Rht-B1 Indel in 56 tetraploid wheat,we found Rht-B1 Indel is not bound to the plant height.5.We cloned Rht-dp allele in 56 tetraploid wheat,and 4 new types of alleles were found and named.We found that Rht-B1^IC12196 is Rht-B1b.
