Tomato is the vegetable crop mainly cultivated in China.It is of great practical significa nce to solve the problem of falling flowers and fruit.The process of abscission includes cell expansion and programmed cell death(PCD).The moisture transport and protect cells turgor are the key to their expansion.Stable H2O2 is involved in the regulation of PCD and regulates programmed death during abscission.Previous studies have reported that different aquaporins are involved in cell expansion and H2O2 transport.Therefore,in order to find out whether aquaporin participates in the abscission process,relevant experiments were carried out,and the key aquaporins Sl PIP1;3,Sl PIP1;5 and Sl TIP1;1 which regulate the abscission,were screened out.1.In this paper,identified the tomato abscission zone specific and highly expressed aquaporin Sl PIP1;3,Sl PIP1;5,Sl PIP2;4,Sl PIP2;5,Sl TIP4;1,Sl TIP1;1 by virus-inducing gene silencing(VIGS),screening out Sl PIP1;3,Sl PIP1;5,Sl TIP1;1 can significantly delay absc ission.2.Due to the similarity of expression patterns of these genes,further double gene silencing and triple genes silencing were performed to obtain p TRV2-Sl PIP1;3Sl PIP1;5,dou ble gene silencing strains and p TRV2-Sl PIP1;3Sl PIP1;5Sl TIP1;1 triple gene silencing plants.For the following conditions: under normal conditions,hydrogen peroxide treatment,the abscission rate under ethylene treatment was statistically found,and plants of p TRV2-Sl T IP1;1 and p TRV2-Sl PIP1;3Sl PIP1;5Sl TIP1;1 were found to delay the induced shedding under hydrogen peroxide conditions.Only p TRV2-Sl PIP1;3Sl PIP1;5Sl TIP1;1 plants can dela y the abscission induced under ethylene conditions,indicating that aquaporin may participate in hydrogen peroxide and ethylene-induced abscissoion pathway through specific members.3.We use transcriptome analysis to further explore the molecular mechanisms of its regulation.The differential genes mainly involve processes such as plant hormone balance,hydrolase,redox reaction,and vesicle transport.These data further indicate that aquaporin not only regulates the transport of water,but also enlarges the cells.It also regulates the condition of the whole cell and the metabolism level of the cells,indicating that aquaporin can affect the cells by regulating the level of redox in the cells.The rate of water absorption and the rate of cell death regulate the occurrence of abscission.
