Grain morphology is one of the important factors affecting yield and quality of rice.In this study,the mutant agl1(Abnormal glume)and Shennong 9816 were used as experimental materials.The structure of the mutant was observed by paraffin section,scanning electron microscopy and scanning electron microscopy.The effects of mutation on important agronomic traits were analyzed,and the mutant gene was cloned.The results are as follows:1.The lemma of mutant agl1 bends inward like a sickle,and the lemma degenerates,partially enclosed by the lemma.Scanning electron microscopy(SEM)showed that the stamens were in a double-slurry state,and the stamens were slightly white.The length of the stamens was shorter than that of the wild type,and the pistils were enlarged.2.The pollen viability of mutant agl1 was significantly decreased,and the pollen was mostly unsaturated and abnormal.Within unit area,the wild-type pollen was aggregated and plump,and the mutant agl1 pollen was dispersed,and the number was significantly less than that of the wild-type.Within unit area,the pollen viability of wild type was 96.4%,and that of mutant agl1 was 88.5%.3.The mutant plant height became shorter and had significant difference.The angle between flag leaves increased by about 20%,and the width of flag leaves increased by about18%.Compared with the wild type,the panicle length of mutant agl1 was relatively short,the panicle weight of mutant agl1 was significantly lower than that of wild type Shennong 9816,the 1000-grain weight of Shennong 9816 was 24.74 g,and the 1000-grain weight of mutant agl1 was only 10.96 g,the difference was significant.The protein content of mutant agl1 was significantly higher than that of wild type,but the amylose and total starch content of mutant agl1 was lower than that of wild type Shennong 9816.4.By map-based cloning,the mutant gene agl1 was located between the molecular markers CH02-140.9 and CH02-143.7 on chromosome 2 with a genetic distance of 2.8c M.Through the analysis and prediction of OFR in the region,it was found that LOC?Os02g56610 coding DUF640 domain gene regulated the development of internal and external lemma in rice.Sequencing of LOC?Os02g56610 revealed that the mutant agl1 had abase substitution(C-T)on the exon,it leads to abnormal translation of amino acids,from glycine to glutamic acid.,which may be the cause of the variation of traits.
