Turnip crinkle virus(TCV)is a member of Crmovirus genus.Its genomic RNA is covered by virus protein coat.Two overlapping proteins,p8 and p9,are expressed from subgenomic RNA1 and are required for cell-to-cell movement and systemic spread of the virus.They also involved in cell-to-cell movement of virus induced RNA silencing.Interaction between protein Atp8 and TCVp8 in Arabidopsis thaliana has been confirmed and analyzed at early of this century.This research is concerned about the interaction of TCVp8 and Nbp8 of Nicotiana benthamiana,and the interacton of TCVp8 with other proteins in N.benthamiana that might involved in cell-to-cell movement of virus induced RNA silencing.Based on yeast two-hybrid analyses,the homolog protein of Atp8 in N.benthamiana-Nbp8,might have an interaction with TCVp8.However,this interaction still need further analyzing.159 TCVp8 interaction proteins of N.benthamiana have been obtained by two-hybrid library screening and conserved domain search.Confering former research,I?ve chosen Nb AT5G37740,Nb SYTA,Nb CSN5 A and Nb WRKY21 for further confirmation and analysis.For the interaction of Nb CSN5 A and TCVp8 hasn?t been confirmed by yeast two-hybrid assays.I will go further on confirmation of the interaction of other proteins and TCVp8.After confirmed the proteins interacting with TCVp8,I will go further on studying the mechanism of cell to cell spreading of RNA silencing.
