Chinese mitten crab(Eriocheir sinensis)is a decidua crustacean.Due to species characteristics of this crab in fighting for food and space,itst survival rate and total production are always low in aquaculture.Therefore,the individual culture of Chinese mitten crab has gradually attracted people's attention in recent years.However,individual culture of Chinese mitten crab has been weakly conducted because of limiting in culture facilities.In this paper,growth rate,molting hormone,fatness and body color changes were firstly analyzed for Chinese mitten crab reared in thecrab individual culture system--"Crab Palace".Then,feed effect and proteomics characters of this crab reared in the "Crab Palace" were investigated from the three kinds of foods.The results of this paper are as follows:1.Observation on the molting and growth of Chinese mitten crab raised in the "Crab Palace"This chapter analyzes the molting and growth characters of Chinese mitten crab reared in the "Crab Palace" for one-molting cycle.The results showed that the average increment rate of body weight of the crab was 33.19%,the average increment rate of carapace length,width and height ranged from 12.12% to 14.97%,the average molting interval was 59.35 d.The average condition factor before molting was 60.84%,and the mean RGB value of body color in back carapace was 109.17,the content of ecdysone hormone(EH)meanly reached 14.18 IU/mL.In the "Crab palace" system,there was no significant correlation(P>0.05)were observed between the growth status,EH content,molting interval before molting and increment rate of the growth traits after molting(P > 0.05).Generally,this study indicated that Chinese mitten crab reared in the “Crab palace” system would start molting when its condition factor reaches 60%,and ecdysone hormone content is more than 14 IU/mL.2.Food effect of Chinese mitten crab reared in " the crab individual culture systemIn order to understand the selectivity and utilization status of different foods in Chinese mitten crab reared in the crab individual culture system--"Crab Palace",this chapter analyzed the growth characteristics,representative digestive enzyme activities of and their correlations among the three kinds of foods(A: water grass + snail;B: water grass;C: snail).The results showed that in the first molting period,the survival rates of the crabs in groups A,B and C were 76.19,41.67 and 69.05%,respectively;the average weight growth rate was 59.06,37.35 and 44.34%,respectively.In the second molting period,the survival rates at the groups A,B and C were 60.94,28.57 and 53.45%,respectively;the molting interval period were 28.70,60.84 and 37.32 d,respectively.The results showed that different kinds of foods had significant effect on molting and growth of Chinese mitten crab.The correlation analysis indicated that there were significantly positive correlation(P < 0.05)between the the trypsin and ?-amylase activities of hepatopancreas,stomach and weight gain rate in the group A,C,while no significant positive correlations(P > 0.05)were found between this two enzyme activity in the two tissues and weight gain rate in the group B.The results showed that Chinese mitten crab in the "Crab Palace" displayed the best growth characters when it was fed with water grass and snail(plant food and animal food).3.Proteomic analysis of Chinese mitten crab reared in " the crab individual culture systemIn order to further understand protein molecular basis of food utilization of Chinese mitten crab reared in the "Crab Palace",this chapter investigated the protein expression difference by using proteomic techniques among the three kinds of foods(A: water grass + snail;B: water grass;C: snail).The results showed that there were 281 significantly differentially expressed proteins between the group A and group B(P<0.05),in which 79 proteins were significantly up-regulated and 202 proteins were significantly down-regulated;There were 364 significantly differentially expressed proteins between the group B and group C(P<0.05),in which 87 proteins were significantly up-regulated and 277 proteins were significantly down-regulated;There were significant differences in the expression of 107 proteins between the group A and group C(P<0.05),in which 83 proteins were significantly up-regulated and 24 proteins were significantly down-regulated.There were significant differences in the expression of 9 proteins among the A,B and C groups,involviing cell structure,fat and energy metabolism.Generally,there was low expression of protein in crab when it fed with water grass.
