Seed Germination Characteristics And Root Segment Reproductive Capacity Of Medical Paeonia Lactiflora Pall.

Belonging to Paeonia species of Paeoniaceae family,Paeonia lactiflora Pall.has been widely used as material of traditional herbal medicine.The dried roots of it contain a variety of active constituents displaying various biological activities such as analgesic,spasmolysis,liver protection,antihypoxic and antimalarial.Thus,there is an increasing demand for cultivars of Paeonia lactiflora Pall.Due to its important pharmacology functions.However,for its characteristic of sterility,the tubers of Paeonia lactiflora Pall.in Zhongjiang of Sichuan Province,China,are used in propagation,resulting in fewer medicinal materials available and lower crop yield and quality.Meantime,the low germination rate resulted from seed dormancy and long growth period of Paeonia lactiflora Pall.in Bozhou of Anhui province,the resources of peony are in short supply in the market.Therefore,there is a need to explore a low-cost asexual cultivation with high propagation coefficient and reveal the factors caused the seed dormancy and the the measures of dormancy-breaking.The results were as follows:1.Firstly,the root segments of Paeonia lactiflora Pall.in Zhongjiang of Sichuan province were soaked with four root promoter with different concentrations to explore the asexual propagation techniques.With different kinds of root promoters,the responses of root emergence rate and root segements growth showed up different in different parts of roots.Considering the costs and effects of root promoter on root emergence and growth as a whole,the promoter B was more suitable for root segement cultivation of Paeonia lactiflora Pall.in Zhongjiang of Sichuan province.2.Secondly,the seed characteristics of water permeability and breathability,the seed embryo development,endogenous inhibitor and hormones contents of Paeonia lactiflora Pall.seed in Bozhou of Anhui province were observed to disclose the reasons lead to seed dormancy.It was observed that the seeds of Paeonia lactiflora Pall.belong to deep combinational dormancy and the hypocotyl and epicotyl of seeds were both in dormant.The husk obstruction to the water uptakes and breathability,morphological embryo hypoplasia,endogenous chemical inhibitors and abnormal hormones levels in seeds of Paeonia lactiflora Pall.might be the reasons of seed dormancy.3.The comprehensive methods to break seed dormancy and facilitate germination were researched.It was revealed tthat there was a need for acomprehensive solution to break the dormancy and promote germination of Paeonia lactiflora Pall.seed.The seeds soaked with 300 mg/L gibberellin acid and storied in sand had the highest rooting rate and strongest rooting ability than those in the control condition.Additionally,cold treatment of the seeds for 4 weeks at 4 ? with roots sprouting accelerated seed dormancy releasing and more rapid and uniforming emergence than that of control.Simultaneously,the seedling of Paeonia lactiflora Pall.seeds with chilling treatment were developed better.
