Spodoptera exigua Hübner is a multi-feeding insect pest that occurs intermittently.Its control is still dominated by the use of chemical pesticides,and bisamide insecticides have become the most prefer agents to control Lepidopteran pests in recent years.In this paper,the differences in growth,development and reproduction of chlorantraniliprole-resistant and susceptible strains of S.exigua were compared,and the differences of molecular mechanisms in fertility between the two strains were discussed.The following results were obtained:1.The bioassay was carried out by the mixed drug method,and the resistant strain?CH-RE?and sensitive strain?CH-SE?were obtained by continuous screening for 6generations by the two-direction screening based on the Leshan population?LS-F?of S.exigua,with the resistace ratios?RR?of 226.69 and 3.72,respectively.2.By anatomizing the female adult ovary of CH-RE and CH-SE strains,the actual egg production and egg production rate of CH-RE strains were significantly lower than those of CH-SE strains,while there were not significant difference in ovarian weight,ovarian tube length and ovariole index.Fluorescence quantitative analysis showed that the vitellogenin genes of CH-RE and CH-SE strains were all expressed on the 5th day after deuteration,and the highest expression values were at 36 h after emergence,but the value of CH-SE strain was significantly higher than that of the CH-RE strain.3.Using the age-stage tow-sex life table analysis method,the differences life parameters of CH-RE and CH-SE strains were compared.The results indicated that the developmental duration,pre-expansion period and adult pre-ovulation period?APOP?of the 3rd instar larvae of CH-RE strains were significantly prolonged compared with the CH-SE strain.The lifespan of male and female adults was significantly increased.And the number of eggs laid was significantly reduced.The intrinsic rate of increase?r?,finite rate of increase???,and the net reproductive rate?R0?were all significantly reduced,and the meean generation time?T?was significantly elongated with a fitness?Rf?of 0.25.4.Using transcriptome sequencing analysis,the differential genes between CH-SE and CH-RE strains were obtained,and 39957 unigenes were spliced together,of which11,891 were annotated,and GO function analysis and KEGG analysis were performed on the unigenes.It was found that differentially expressed unigenes were significantly enriched in metabolic pathways.Eight up-regulated genes related to the reproduction and resistance in transcriptome data were verified by real-time PCR,and the results were basically consistent with the expression of transcriptome data.
