Vertebrate retina is a model system for the development and function of central nervous system.The development of nervous system is accompanied by complicated genetic regulation.Long noncoding RNAs(lncRNAs)have close relationships with the development and function of the central nervous system(CNS).Previous studies on the retina focused mostly on its structure and the diseases that affect it.However,the changes that occur in m RNA and lncRNA expression during the process of retinal development at pre-and postnatal stages have remained unclear.In this study,pigs were used as animal models,and transcriptome sequencing was used to study the changes of retina between prenatal(piglets,pregnancy for 114 days)and postnatal(2 days after birth).The results as follows:(1)In current study,5 samples from prenatal and postnatal group of retina were sequenced by RNA-seq and produced in total 59.69 GB of Raw data.Compare with reference datas,average 77.9% of clean reads from those clean data could be mapped to the porcine reference genome.(2)There were 25,323 of mRNA transcripts and 15,428 of lncRNA transcripts identified from 5 sequenced libraries.The identified lncRNA have lower protein coding potential,less exon numbers and lower FPKM than identified m RNA in current study.(3)There were 99 differential expressed m RNAs and 317 differential expressed lncRNAs through difference analysis.BMPER act as one of the most different m RNAs which could play an important role in retinal development.(4)From the functional analysis of highly expressed lncRNAs,their potential functions about correlated m RNAs could be significantly enriched in some biological process,such as the photoreceptor inner segment,sensory perception of light stimulus and the dorsal ventral axis formation.(5)From the functional analysis of differential expressed lncRNAs,their potential functions could be enriched in synapsis and retinal development.Lnc RNA TU69849 and TU20386 have lower FPKM in postnatal.(6)From the functional analysis of differential expressed miRNAs,miR-26a-5p could affect retinal development through RANBP9 of piglet.
