In this study,Favorita and Ba Shushihao were choosed as the potato experimental materials.Through pot experiments,Eight physiological indexes were measured with four treatment levels of potassium sulfate and potassium.In addition,the potassium related genes of potato were cloned and bioinformatics analyzed.The main results were as follows:(1)In the design of potassium application(CK as the control,K2,K3,K4 were potassium sulphate treatments,pure potassium content:45kg/hm~2?90kg/hm~2?180kg/hm~2.K5,K6,K7were potassium chloride treatments,pure potassium content:45kg/hm~2?90kg/hm~2?180kg/hm~2).Potassium sulfate for the promotion of two kinds of potato plant height growth was better than that of potassium chloride.With the application of potassium sulfate,K4treatment was the most significant increase in potato plant height.With the application of potassium chloride,the height of potato plant height increased most significantly by K7treatment.(2)During the treatment of K4 and K7,the yield of potato tuber reached the maximum value of potassium sulfate and potassium chloride.There was no significant difference in the effect of potassium sulfate and potassium chloride on the yield of potato per plant.(3)When potassium sulfate was applied,the starch content of Favorita increased 2.2%.and the starch content of Ba Shushihao increased 2.8%.When potassium chloride was applied,the starch content of Favorita increased 2%and the starch content of Shushihao increased2.3%.Potassium sulfate was more conducive to potato starch accumulation than potassium chloride in the same pure potassium content.(4)With increasing potassium application,the content of reducing sugar in potato tubers showed a decreasing trend.The reducing sugar content of potato reached the lowest value with K4 and K7 treatments respectively.(5)During the potato tuber formation period,the chlorophyll content in leaves increased with increasing potassium application.Under the same pure potassium content,the chlorophyll content with potassium chloride treatment was higher than potassium sulfate treatment.(6)When the content of pure potassium was less than 180kg/hm~2,the soluble protein content increased with increasing potassium application.The soluble protein content in potato tuber with potassium sulfate application was higher than potassium chloride application.(7)During potato tuber formation stage,potassium content in potato leaves increased gradually with increasing potassium application.When the content of pure potassium was higher than 45kg/hm~2,potassium chloride application was more helpful to accumulate potassium than potassium sulfate application.And there was no significant difference in potassium content of tubers.(8)Five genes homologous of Arabidopsis thaliana were identified in potato genome by comparative genomics.These five genes were CIPK6,CIPK14,CIPK9,CBL4 and NKT1respectively.The gene which had homology with Arabidopsis CIPK14 was named StCIPK14.They have the highest homology with Arabidopsis AtCIPK5,AtCIPK14,AtCIPK6,AtCBL4and AtAKT1.(9)CIPK6,CIPK14,CIPK9 and CBL4 genes were stable proteins.NKT1 was unstable proteins.All the proteins were hydrophilic proteins and they were not secretory proteins.Subcellular localization prediction indicated that CIPK6,CIPK14,CIPK9 and CBL4 might be located in cytoplasm and NKT1 might be located in endoplasmic reticulum.(10)The results of functional conservative domain analysis showed that the proteins encoded by CIPK6,CIPK14 and CIPK9 genes had"cd12195"structure.The protein encoded by CBL4 gene had"cd00051"structure.The protein encoded by NKT1 gene had"PLN03192"structure.
