Objective: Enteric glial cell(EGC)is derived from the neuroectoderm and is one of the main members of the intestinal nervous system.EGCs mainly has the functions of nutrition and supporting neurons and can regulate the occurrence and development of intestinal inflammation to a certain extent.Previous studies on EGC mainly focused on the intestinal inflammatory disease of mammalian mice,and there was a lack of research data on chicken intestinal EGC.In the chicken enteritis disease,necrotizing enteritis can infect the chicken each day age stage,the mortality rate is high,after come on,can cause huge loss to breeding industry.In this paper,xinjiang yellow-feathered broiler chickens were used as research animals to establish A model of necrotizing enteritis disease in chickens,and the morphological structure and distribution of EGC in the intestinal tract of necrotic enteritis chickens were studied in detail,as well as the expression of GDNF,neuropeptide A and SOX9 in EGC secretion factors in the entire intestinal tract,and their functions were analyzed.It lays a theoretical foundation for further study of its physiological function and immune function,and provides scientific basis for breeding,disease prevention and treatment of chickens.Methods: in this study,80 healthy yellow-feathered broilers with a similar body weight of 50 days were selected,40 males and 40 females respectively,and they were divided into control group and experimental group.In the experimental group,necrotizing enteritis was induced by artificial infection of clostridium weilensis type A.40 yellow-feathered broilers in the experimental group were subjected to oral oral administration of clostridium weilensis 15 x 108cfu/ chickens for 10 consecutive days,until the chickens were slaughtered immediately after clinical symptoms.Some of the samples were fixed in 2.5% glutaraldehyde solution,which was used for making electron microscopy materials.Another part was fixed in helly solution for improved Holzer staining;The third part was fixed in 10% neutral formaldehyde solution for immunohistochemical tests.The control group was the same as the experimental group.Results: application of tem and study on the method of modified Holzer dyeing necrotizing enteritis in enteric glial cells morphological structure characteristics and distribution law of electron microscope to its ultrastructure observations for necrotizing enteritis in experimental intestinal EGC cytoplasmic vacuoles increase,cytoplasm,uneven distribution of decreased significantly and the clearance between two adjacent cells increases,fracture.The results of optical microscopy showed that the EGC in the control group was widely distributed in the mucosa,submucosa and muscle layer of the chicken intestinal tract,with a small amount of distribution between the mucosa epithelium.Immunohistochemical staining was used to observe,and the results showed that the neuropeptide A in the intestinal tract of necrotic enterocolitis chickens mainly expressed EGCs in the submucosal plexus and intramuscular nerve plexuses.The glial neurotrophic factor(GDNF)secreted by intestinal glial cells is mainly expressed in the mucosal epithelium,the small intestinal gland and its surrounding vessels in necrotic enterocolitis chickens.SOX9 secreted by intestinal glial cells is mainly expressed in the mucosal epithelium and lamina propria in the intestinal tract of necrotic enterocolitis chickens.Conclusion: the morphological changes and quantity of intestinal glial cells in necrotic enterocolitis chickens were reduced.Neuropeptide A,GDNF and SOX9 secreted by intestinal glial cells were mainly expressed in the mucosa,submucosa and intramuscular plexus in the intestinal tract of necrotic enterocolitis chickens,and the expression was reduced compared with the control group.
