Paeonia Suffruticosa is a genus of peony and peony,native to China.The peony is gorgeous and magnificent,and the flowers are large and fragrant.Peony is a traditional Chinese ornamental plant with high ornamental,medicinal and oil value.It is a good garden landscaping plant and cash crop.Plant pigments are mainly composed of flavonoids,carotenoids,and betaine pigments.The content and composition of flavonoids are completely different in petals of different colors of the same species.In this experiment,three color crops of 9 peony varieties were used as experimental materials,and the pigment content of different color peony petals was analyzed by thin layer chromatography,ultraviolet-visible spectrophotometry and high performance liquid chromatography.To determine the reason for the different coloration of peony petals,the expression levels of MYB and bHLH and structural genes PAL,CHS,CHI,F3 H,DFR,ANS,UF3 GT and UF5 GT in the flavonoid biosynthesis pathway were also analyzed.It provides a theoretical reference for the cultivation of new varieties of peony,the increase of the beauty of garden landscape and the selection of peony varieties with more economic value.The results showed that all peony petals contained flavonol and flavanone substances,and pink and red peony petals contained cyanidin-3,5-diglucoside,peony--3,5-diglucoside,There are four anthocyanins of cyanidin-3-glucoside and peonidin-3-glucoside,but no anthocyanins are detected in the petals of white peony.The color of the flower color was deeper than the increase of total flavonoids.For chlorophyll and carotenoids,only traces of chlorophyll were detected in the pink variety 'Coral Taiwan',and chlorophyll was not detected in other varieties of peony petals.Carotenoids were also detected only in the red varieties 'Oolong' and 'Morunn',and the content was extremely low,which was not detected in other varieties.The methanol extracts of peony petals of different color systems were analyzed by HPLC-DAD and the results were calculated.It is concluded that the total anthocyanin content of 'Xueying Peach Blossom' 'Silver Red' on ' Caotaitai' 'Wei Zi'' Wulong Zhisheng''Morun Juelun' is: 4.433mg/g,17.133 mg/g,39.221 mg/g,138.352 mg/g,115.142 mg/g,254.313 mg/g.In addition to anthocyanins,two co-pigments,quercetin and hibiscus,were detected in the purple peony,which aided in coloration and deepened the petals.Real-time quantitative PCR analysis of the petals of different color peony trees.The expression levels of MYB and bHLH and the structural genes PAL,CHS,CHI,F3 H,DFR,ANS,UF3 GT and UF5 GT in the flavonoid biosynthesis pathway were analyzed.The structural genes CHS,F3 H,PAL,UF5 GT and transcription factor bHLH3 were higher in the white peony petals than in the pink and purple lines;the structural gene DFR was the highest in the pink peony petals;the structural genes CHI,ANS,UF3 GT and the transcription factor MYB22 were most expressed in the petals of purple peony.
