Zucchini yellow mosaic virus(ZYMV),a potyvirus,has been causing increasingly serious damage to cucumber(Cucumis sativus L.)in recent years.ZYMV can interfere the growth and reduce the yield as well as the quality of cucumber.It is necessary to develop ZYMV resistance because there are no effective chemical substances to control ZYMV.ZYMV Z5-1was used as virus source and candidate gene was sequenced in order to identify ZYMV resistance of different cucumber materials.The results show that cucumber lines SA02,SA06,S94,S1003,WI7230 and TMG-1is resistant to ZYMV.Gene-internal marker,InDel-zym,were designed based on the sequence of the ZYMV-resistant candidate gene Csa6M152960.1.ZYMV resistant gene in TMG-1,zymTMG-1,was introduced into a ZYMV susceptible cucumber line SA0422 through backcross breeding so as to acquire a cucumber line which is resistant to ZYMV and has excellent quality characters.During the backcross breeding,foreground and background selections were conducted in backcross population.SA0422 possessing ZYMV resistant gene zymTMG-1 was developed within three generations of backcross,laying a solid foundation for breeding new ZYMV-resistant cucumber lines. |
