With the development of economy,the requirements of livestock and poultry production performance are higher and higher.Existing studies have found that FSH and LH can improve animal reproductive performance,reduce follicular apoptosis and follicular atresia.mirna is closely related to animal diseases and apoptosis.therefore,by adjusting FSH and LH,mirna expression can be changed,and finally animal production performance can be improved,which provides possibility for improving livestock and poultry productivity.In this study,chicken follicular granulocyte cells in vitro different concentrations(0,5,10 ng/mL)FSH or(0,2,4 ng/mL)LH were used to treat,and cells were collected after 0,24,and 48 H.The expression levels of miR-21,miR-181 a,miR-125 b,miR-let-7a were detected by qRT-PCR to further analyze the effects of FSH and LH on the expression of four miRNAs associated with apoptosis.The results are as follows:(1)The effect of FSH on the expression level of four kinds of miRNA: compared with the control group(0,24,48h),after adding FSH(0,5,10ng/mL),the expression of miR-21 was significantly different(P<0.01).At the same time,the higher the concentration of FSH,the higher the level of miR-21 expression.After adding FSH(0,5,and 10ng/mL),the expression of miR-181 a,miR-125 b and miR-let-7a were all significantly different(P<0.05).At the same time,the addition of 10ng/mL FSH to the group adding 5ng/mL FSH,the 3 miRNA expression levels decreased,indicating that the different concentrations of FSH had different effects on the expression.(2)The effect of LH on the expression level of four kinds of miRNA: compared with the control group,with time change(0,24,48h),after adding LH(0,2,4ng/mL),miR-21,miR-let-7a expression after 24 h was not significantly different(P>0.05),and 2 miRNA expressions after 48 h were significantly different(P<0.05).At the same time(24h),after adding 2ng/mL LH,the expression level of the 2 miRNA was higher than that of the 4ng/mL LH group,but 48 h showed the opposite change.After adding LH(0,2,and 4ng/mL),the expression of miR-125 b and miR-181 a were significantly different(P<0.05),at the same time(24h),and 4ng/mL LH was compared to the group adding 2ng/mL FSH,and miR-181 a expression level decreased,but after 48 h was the opposite change.(3)At the same time,under different concentrations of FSH,except for miR-21,the expressions of miR-125 b,miR-181 a and miR-let-7a were significantly different(P<0.05).The 4 miRNA expressions were 5ng/mL FSH higher than 10ng/mL FSH,and the 4 miRNA changes were the same,indicating that different concentrations of FSH have different effects on the expression of 4 kinds of miRNA in chicken follicle granulosa cells.(4)At the same time,under different concentrations of LH,except for miR-21 and miR-let-7a,the expressions of miR-125 b and miR-181 a were significantly different(P<0.05).The 4 miRNA expressions were 4ng/mL LH higher than 2ng/mL FSH,and the 4 miRNA changes were the same,indicating that different concentrations of LH have different effects on the expression of 4 kinds of miRNA in chicken follicle granulosa cells.
