| Researching the effects of peony seed oil on Schizothorax prenanti immune function.on the one hand,it provides a theoretical basis for the suitable plant fat source for the fish feed of Schizothorax prenanti.On the other hand,it provides the basis for the application of peony seed oil as functional food.After feeding the peony seed oil for 8 weeks,Through the determination of immune related indicators: C3,IgM,CRP levels,GSH-Px activity;antioxidant system-associated indicators: NO,iNOS,MDA levels,SOD,GSH-Px,LZM activity;nutritional status related indexes:ALB,HDL-C,LDL-C,T-CHO,TG,TP levels.The expression of immune related genes tlr5,tlr22,myd88,il8,,il10,tgf-β1,tnf-α,traf6,cd40,pkr,nf-kbp100,β2-integrin,irf7,g6 pd were detected in the head kidney,spleen,liver and mid-gut by fluorescent quantitative PCR.The effects of peony seed oil on immune function at 6h,24 h,72h and 168 h Schizothorax prenanti infected with Aeromonas hydrophila were studied.The results are as follows:(1)Peony seed oil increased the content of fat,ash and protein in the muscles of Schizothorax prenanti,but there was no significant difference in water content.The specific growth rate and weight gain rate of Schizothorax prenanti were improved,and the fasting index and the length ratio of the intestines were reduced,and the other indexes were not significantly different.(2)Compared with the control group,in terms of immune indicators,peony seed oil significantly increased Schizothorax prenanti serum C3,IgM levels,there was no significant difference between ACH50 and CRP;In terms of antioxidant indexes,peony seed oil significantly increased Schizothorax prenanti serum GSH-Px,SOD activity,reduced NO,MDA activity;In terms of nutrition indicators,peony seed oil significantly increased Schizothorax prenanti serum TG levels,reduced TP,T-CHO levels,there were no significant differences in LDL-C,HDL-C,ALB,lactic acid and CK.(3)After infection of Aeromonas hydrophila in Schizothorax prenanti,at 6hours,peony seed oil increased ACH50,C3,IgM,MDA,NO,iNOS,LZM levels.As the infection time,at 24 hours,peony seed oil increased C3,ALB,iNOS,MDA levels.At 72 hours,peony seed oil increased CRP levels.IgM levels higher than control group during the infection time.The rest of indicators are not significant compared with control group.It indicated that Aeromonas hydrophila infection stimulated the immune response of Schizothorax prenanti,and feeding of peony seed oil increased the immune response in serum,but the effect on serum antioxidant indexes and nutritional indicators were not significant.(4)Compared with the control group,peony seed oil significantly increased the relative expression of pkr,nf-kbp100,traf6,tgf-β1 and tlr5 in head kidney,the relative expression ofcd40,traf6,tgf-β1,tlr5 and tlr22 in spleen,the relative expression ofcd40,il10,myd88 and g6 pd in liver,the relative expression ofcd40,nf-kbp100,il10,tnf-α and tlr22 in mid-gut.(5)After Aeromonas hydrophila infection,peony seed oil rapidly increases the relative expression of immune genes in Schizothorax prenanti.As the infection time,the body gradually restore balance.But at 168 hours,there is a significant expression of immune genes in head kidney,spleen and liver.Therefore,we can draw the following conclusions:Peony seed oil can improve Schizothorax prenanti immunity.After Aeromonas hydrophila infection,peony seed oil can adjust serum related immune protein and the relative expression of immune genes in tissues.It enhances the Schizothorax prenanti against pathogens-Aeromonas hydrophila to keep the fish healthy growth. |
