Puberty is a period of transition in which adolescents' reach sexual maturity.It is the period when the females first incur estrus and ovulation.The previous studies have evaluated this process mostly in rodents and primates.The current results report partial outcomes on mechanism involved during puberty in ewes.We explored the effect of DNA methylation on the expression of key genes in the eustachian initiation of the aculate in the hypothalamus using the dual-binding method of F-MSAP and BSP.1.A total of 2,409 bands were found during the early stages towards with concentration at 21-309 bp.The premethylation model was predominantly in the preand prepubertal stages,and the semi-methylation model was predominant in the stage of prepuberalization.The hypothalamic methylation level was 34.20% at the initiation of puberty,which was significantly lower than that of the adjacent puberty period(61.96%),P<0.05.It shows that there is a significant difference in the patterns and levels of DNA methylation before and after initiation of eustachian ejaculation.2.During puberty,NKB gene is expressed in the hypothalamus of ewes in small quantity,it gradually increases during the subsequent stages of puberty.3.During the initiation stage of puberty,the overall methylation level of the NKB promoter region was significantly reduced,and the overall methylation level during the puberty period was not significantly different from that of the adjacent puberty stages;the specific site of the promoter region(-1261)-1254 and-836 bp methylation levels were significantly reduced.This study shows that the methylation of genomic DNA and the methylation of NKB gene are important factors influencing the initiation of eclampsia in ewes.
