The Oocyst Wall Protein TgOWP8 Of Toxoplasma Gondii And Application In Detection Of Antibodies In Serum Of Pigs And Chickens

Toxoplasma gondii is an opportunistic protozoan which can be widespread parasitization in a variety of animal cells.The main route of transmission is congenital transmission and acquired sexual transmission.Humans or animals can acquire acquired infections by eating foods contaminated by oocysts and cysts.Therefore,Toxoplasma gondii oocysts and cysts play a key role in the transmission of this parasite.Studies have shown that oocyst-induced Toxoplasma gondii infection may be more clinically meaningful than the infection obtained by tissue cysts.TgOWP8 is a newly identified Toxoplasma gondii oocyst wall protein,but its specific studies are still few.In this study,TgOWP8 was studied and based on the expressed and purified TgOWP8 recombinant protein,the coating concentration was determined at a dilution of primary antibody of 1:50,a secondary antibody dilution of 1:20000,and a color development time of 25 min.Gradient dilution was performed to determine the optimal ELISA coating concentration of TgOWP8 at 5?g/ml;and ELISA assays were performed using tachyzoites(TLA)and TgOWP8 on porcine tachyzoite positive and negative serum,farm pigs and chicken serum.The ELISA test results of the two proteins were compared and analyzed.The results of swine tachyzoan serum test showed that the positive rate of positive serum TLA was 100%(15/15),and the positive rate of TgOWP8 was 93%(14/15).There was no difference in sera,indicating that TgOWP8 protein could better distinguish Toxoplasma gondii infections;the pig farm serum test results showed that the positive rate of TLA was 16.7%(15/90),and the positive rate of TgOWP8 was 12.2%(11/90);The chicken serum test results showed that the positive rate of TLA was 10.4%(10/96),and the positive rate of TgOWP8 was 13.5%(13/96),indicating that the infection pathway of pigs and chickens infected with Toxoplasma gondii was mainly through oocyst infection.Positive sera were also verified by Westing blot.It was demonstrated that TgOWP8 can be used as an indicator of Toxoplasma infection in oocysts.
