Pseudorabies virus is a kind of acute infectious disease caused by pseudorabies virus,which is characterized by fever of piglets,encephalomyelitis and abortion of pregnant sows.It is one of the major infectious diseases that seriously threaten the breeding industry.In recent years,with the emergence of various strains of pseudorabies virus,the vaccine has little limitation on the prevention and treatment of pseudorabies.Therefore,it is necessary to find and develop effective antiviral drugs to control pseudorabies.In recent years,quinolones have made some achievements in antiviral research,and a series of quinolones with herpes viral activity have been reported at home and abroad.Levofloxacin is one of quinolones.Although the state has explicitly prohibited the use of levofloxacin as a veterinary antibiotic,its antiviral effect needs further study and utilization.Many domestic and foreign literatures have reported the antiviral effect of levofloxacin,such as on hepatitis B virus.Inhibition,treatment of respiratory syncytial virus infection,etc.This paper explores the effect of levofloxacin on the proliferation of PRV,and studies the anti-PRV activity of levofloxacin in mice and cells in vitro and in vivo.It is of great significance to provide a new way for the clinical prevention and treatment of PRV by levofloxacin and to explore new resources of antiviral drugs.1.BHK-21 cells were cultured on 96-well plate and inoculated with multiple diluted virus.The pathological changes of the cells were observed.The viral titer of PRV-XJ strain was 106.79 TCID50/0.1 mL.2.The method of culturing cells again and inoculating multiple to dilute the virus solution was the same as the first experiment.However,levofloxacin with different multiple dilutions was added to the experiment at the same time.The virus titer was 106TCID50/0.1 mL.The results showed that the titer of virus decreased and the proliferation of virus was affected.It was proved that levofloxacin could inhibit the virus proliferation to some extent,and the virus solution was diluted at 10-4-10-6 and the drug was 10-20 times diluted at 18-30 H.When released,the inhibitory effect of drugs on virus proliferation was the most obvious.3.Starting from the mice in vivo,the mice were injected with different multiples of virus solution.The incidence of the mice was observed.The viral titer of PRV-XJ strain was 103.16.16 LD50/0.1mL.4.The experiment continued to select mice to repeat the attack test in groups,but after48 hours,the mice were injected with levofloxacin diluted by different times.The experiment showed that the number of mice died was reduced,and the days of death were delayed by about 1-2 days.The titer of the virus was 102.48.48 LD50/0.1 mL under this condition,The results showed that the titer of virus decreased and the proliferation of virus was affected.showed that levofloxacin could inhibit the proliferation of PRV in mice,and the inhibitory effect of levofloxacin on the proliferation of PRV was the most significant when the virus solution was diluted at 10-1-10-3 and 10-fold.In conclusion,levofloxacin can inhibit the proliferation of PRV in vitro and in vivo.To provide experimental model and data basis for the follow-up study of PRV antiviral drugs,to provide new ideas for the study of new anti-PRV drugs,and to open up new ways and directions for the treatment of pseudorabies.
