Objective:To study the effect of diallyl disulfide(DADS)against Marek virus(Marek’s Disease Virus,MDV)and use this active ingredient as a parent to develop novel antiviral cancer chemotherapy drugs for the development and treatment of Marek’s disease which provide a theoretical basis for the development of new drugs for the treatment of Marek’s disease.Methods:(1)To establish the model of chicken embryo fibroblast,MDV amplification,collection and titer determination,and the determination of DADS safety concentration.(2)A model was established with healthy chicks as experimental animals.A standard curve was prepared by fluorescence quantitative PCR and the number of copies of the virus was detected.(3)75 healthy 1-day-old chicks were randomly divided into 5 groups(15 in each group),which were set as blank group,control group,and test group(in which DADS in the test group was 750μmoL/mL as the high-dose group,DADS:500μmoL/mL for the drug concentration group,DADS:250μmoL/mL for the drug low concentration group),and clinical trials were conducted on the test animals.At the age of 40 days,the biochemical indexes of the experimental group,the control group and the blank group were tested,and the histological sections of the related organs were made.Pathological changes were calculated,and the copy number of MDV was calculated.Results:(1)The titer of the virus in this test was 3.0×10~5(pfu/mL),which means that it contained 3.0×10~5 virulent virus particles per 1 mL of the virus solution.(2)The optimum concentration of DADS is 100μmoL/mL,150μmoL/mL is a high concentration,and 50μmoL/mL is a low concentration.(3)The results of biochemical indicators showed that the contents of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in the chicken serum at the age of 40 days were normal level in the blank group.Compared with the control group,the AST and ALT of the experimental group under the different DADS concentrations were significantly lower than the control group(P<0.05),and the DADS middle concentration group was significantly lower than the DADS low concentration group(P<0.05).The DADS high concentration group Significantly lower than the DADS middle concentration group(P<0.05),indicating that under the DADS concentration of250-750μmoL/mL,the liver function regulation and protection has a significant role in promoting,and with the increase of DADS concentration,the promotion effect More obvious.(4)Liver tissue section results showed that the control group and the low-concentration group were experimental animals:the necrosis of liver cells was predominant,and the division of the tumor cells was obvious;the hepatocytes of the experimental animals in the high-and medium-density groups were in early stage of necrosis,and the inflammatory cells were slight.Infiltration,some nuclei have pyknotic phenomenon.The hepatic lobule structure,hepatocyte arrangement,and hepatocyte structure were normal in the liver of the blank group.(5)By establishing the standard curve of fluorescence quantitative PCR,the MDV copy number=10^(35.746+3.7246-CT)/3.7246)was obtained.The control group average CT value was 25.54,the average copy number is 5.62×10~6;the high concentration group average CT value is 28.68,the average copy number is 8.23×10~5;the concentration of group average CT of 27.47,the average copy number is 1.73×10~6;the low concentration group the CT value 26.04,copy number average value is 4.06×10~6.After analysis,the number of MDV copies in the three groups of experimental groups with different concentrations of DADS were significantly lower than that of the control group(P<0.05),indicating that DADS could inhibit the amplification of MDV in chickens.Conclusion:The results of serum biochemistry,liver histopathology,and virus copy number analysis showed that DADS had a significant inhibitory effect against the amplification of Marek’s disease virus in chickens;in terms of clinical manifestation,DADS has obvious effect on relieving Marek’s disease.However,the ways and means of the action of DADS on the virus,the optimal dosage forms and dosage of the drugs need to continue to be studied.
