The breeding and planting of transgenic insect-resistant cotton effectively controlled the hazards of cotton bollworms and other pests.The yield and benefits of cotton have been improved and played an important role in the production of cotton.At present,with the continuous development of insect-resistant cotton varieties,not only the breeding direction of insect-resistant cotton needs to be adjusted,but also the cultivation techniques suitable for insect-resistant cotton varieties should be improved.Based on the previous research of the research group,this study selected CCRI89 and SCRC28 two transgenic insect-resistant cotton varieties to explore suitable cultivation methods for transgenic cotton varieties,to analyze its effects on cotton morphology,yield and quality,in order to promote the promotion of transgenic cotton varieties.Cotton is an important fiber and oil crop.In the production,the production of cotton is greatly reduced due to the occurrence of verticillium,and the quality is seriously degraded.Practice has proved that cultivating cotton resistant varieties is the most economical,efficient and environmentally friendly method for controlling verticillium wilt.In this laboratory,the full-length cDNA of cotton VIP1 gene was cloned by homologous sequence method.Functional analysis showed that cotton VIP1 gene has positive regulation against Verticillium wilt traits.To this end,this study used 18 elite cotton germplasms as materials to identify its disease resistance using VIP1 gene,and analyzed the relationship between the gene and cotton-related phenotypic traits,and explored the effects of cotton resistance genes on yield and fiber quality.The main results are as follows:1.CCRI89 has better yield and fiber quality when exposed to the open field;CCRI89has stable yield and fiber quality during simplified pruning;CCRI89 is stable in yield and fiber quality at a density of 90,000 plants·hm-2.2.Based on the influence of various factors on CCRI89,the results showed that CCRI89 is seeded at a density of 90,000 plants/hm2,and the yield and fiber quality are stable when the simplified pruning treatment was carried out.3.The VIP1 gene was used to identify the disease resistance of 18 elite cotton germplasms,and the allelic differences between the gene sequences were analyzed.It was found that there were 6 SNP loci and 1 InDel locus.There are 4 mutation sites in the 4exons of the gene,1 mutation site upstream of the initiation codon,and 2 mutation sites downstream of the stop codon.Moreover,these mutation sites have a significant effect on the phenotypic traits such as the verticillium wilt index,plant height,boll weight and clothing score.
