Common bean?Phaseolus vulgaris L.? is one of important side crops in China and around the world,it features various cultivars with colorful appearance,relative small genome size,short life cycle and a close phylogentic relationship with soybean.Therefore,common bean is an important genetic resource for studying gene function in legume.In this study,seeds of golden hook?Huang-Jin-Gou? were exposed to 600 Co?radiation at a dosage of 200 Gy,the phenotypic observation was performed at M3 generation.It was observed that there were obvious variations in varous triats.Among 3500 M3 lines,each of which was descended from a single M2 plants,607 lines were showed obvious phenotypic changes in growth period?flowering and to maturity?,leaf,plant type,fertility,seed coat color when compared with wild type.33 lines displayed a segregation of wild and mutantion phenotypes,which was in agreement with median 3:1 segregation ratio,indicating those mutations are generally controlled by single recessive gene.We took 100 seed weight and seed size as examples to estimate the phenotypic variations of quantitative traits,and found that mutant lines displayed a large phenotypic variations in these three measured traits,100 seed weight,seed length,and seed width when compared with the wild type of 'golden hook'.It showed statistically significant differences in 100 seed weight,seed length,and seed width between mutants and wild type of 'golden hook'.Some mutations were phenotypically confirmed in the M4 generation.Of total 30 plants of A230 mutant line,7 plants showed seedless pod sterile?SLPS?,in agreement with median 3:1 segregation ratio.A St230 F2 population was obtained by cross between PI60234 as ovule parent and A230 wild phenotype?assuming 2/3 of which were carrying heterozygous SLPS gene? as pollen parent.Of137 plants of St230 F2 population,35 plants displayed SLPS.48 plants of DNA were selected QTL mapping of mutation gene using BARCBean6K3 BeadChip technology.1545 polymorphic markers were scored to build linkage map,the candidate gene was mapped to a range between 990752 and 3463964bp on chr03?about 2.5cM?,explaining 75.81% of total phenotypic variation.
