Apostichopus japonicus is an important marine animal that is loved by our people.However,with the continuous expansion of the Apostichopus japonicus breeding industry,problems such as the decline of germplasm and uneven growth performance have arisen.In order to explore the growth and germplasm improvement of Apostichopus japonicus,hybrid breeding is one of the main methods.It can not only produce new varieties by combining with the superior traits of the parents,but also serve as a reference for subsequent genetic structure analysis.This experiment is divided into three parts.In the first part,in order to explore the molecular differences of pure thorns involved in hybrid thorns,this experiment used Russian Apostichopus japonicus(R)and Chinese Apostichopus japonicus(C)as parents to construct a hybrid family B(R♂×C♀),pure family C(R♂×R♀)and D(C♂×C♀)three families were cultured under the same temperature,salinity,pH and water conditions as follow-up experimental materials.In the second part,a transcriptome library of body wall tissues of B,C and D families at 18 months old was constructed.Through bioinformatics analysis,the results were compared and analyzed.There were 5,330 differential genes in group B and group C of Apostichopus japonicus.There were 6,055 differential genes in the differential genes of group B and group D of Apostichopus japonicus,and 5,875 differential genes in group C and group D of Apostichopus japonicus.Of the 5,330 differential genes in the B and C groups of the Apostichopus japonicus,2,378 genes were significantly up-regulated in the C group,while 2,952 genes were significantly up-regulated in the B group.A total of 382 differential genes were enriched into 169 pathways in the enrichment analysis of differentially expressed genes in Groups B and C.Among them,the number of differentially expressed genes enriched by endocytosis was the highest,at 28.In the third part,the hybrid progeny and the pure progeny have different degrees of phenotypic differences.The heterosis is obvious.The molecular mechanism that affects this advantage is unclear.In the existing transcriptome data of the laboratory,six growth-related genes were selected,and four tissues including body wall,lameness,tube foot and longitudinal muscle at 12 months and 18 months of age were used to study the different tissues of the Apostichopus japonicus.The distribution and its expression between different tissues in different experimental groups.The results showed that the six genes were expressed in different degrees in body wall,lameness,tube foot and longitudinal muscle at 12 months and 18 months of age.The expression levels of Fgfrbp2,Igf2,1,Igf2,2,Megf6,4,and Megf11,3 in the tube foot tissues of 12-month-old family C were significantly higher than those in other tissues.The expression levels of Igf2,1,Igf2,2,and Megf6,1 genes in the 18-month-old longitudinal muscle tissues of the three families were significantly higher than those in other tissues.The expression of the body wall tissue was significantly higher than that of the family C in the 12-month-old family,and the expression of the family C was significantly higher than that of the family D;the expression of the family B and the family C of the 18-month-old was higher than that of the family D.There is no obvious regularity in the expression between family B and family C.
