Carposina sasakii Matsumura is a kind of fruit boring pest that seriously endangers the fruit tree industry in Northern China.It is distributed in other provinces except Tibet,Hainan and Xinjiang provinces in China,which mainly harm apple and jujube.In order to explore the behavioral regulation technology of C.sasakii Matsumura,it is necessary to study the olfactory system of C.sasakii in detail,and to excavate the olfactory genes of C.sasakii.The expression of olfactory related genes was studied by constructing the transcriptome database of antennae at different developmental stages and bioinformatics analysis.The differentially expressed genes related to olfactory were screened.The results were as follows(1)Candidate olfactory related gene sequences in transcriptome databases,According to the annotation,43 candidate genes of odorant binding proteins(OBPs),3 candidate genes of pheromone binding proteins(PBPs),3 candidate genes of general odorant binding proteins(GOBPs),45 candidate genes of chemosensory proteins(CSPs),75 candidate genes of odorant receptors(ORs),3 candidate genes of f olfactory co-receptor(ORCO),14 candidate genes of ionotropic receptors(IRs),88 candidate genes of gustatory receptor(GRs),6 candidate genes for odorant degrading enzyme(ODEs)and 13 candidate genes of pheromone degrading enzyme(PDEs).Among them 24 differentially expressed OBPs,3 differentially expressed PBPs,3 differentially expressed GOBPs,15 differentially expressed CSPs,21 differentially expressed ORs,1 differentially expressed ORCO,4 differentially expressed IRs,20 differentially expressed GRs,3 differentially expressed ODEs and 3 differentially expressed PDEs.(2)The expression of olfactory related key genes in male and female C.sasakii was significantly different between different developmental stages,which was related to the behavioral characteristics of male and female moth at different developmental stages.The expression of odorant receptors(ORs)and gustatory receptors(GRs)was mainly related to gender.The expression of chemosensory proteins(CSPs)is mainly related to sexual maturity of C.sasakii,and the expression of CSPs during sexual maturity,which is related to the expression of ORs and GRs,and the expression of CSPs is mainly related to sexual maturity of insects,and the expression of GRs is related to sex.It is speculated that CSPs is not mature during the eclosion of adult stage,and the expression of odorant binding proteins(OBPs)is mainly related to the copulation.It is inferred that the OBPs regulates insect behavior through different states of the adult insect.(3)Both OBP4 and OBP17 were expressed in male C.sasakii at 0 h and 24 h after eclosion.OBP17 was expressed both in eclosion 0 h stage and eclosion 24 h stage of male C.sasakii,and expressed OBP17 in female eclosion 0 h stage and female eclosion 24 h stage as well.OBP4 was only expressed in mated females.The expression of OBP4 in female was higher than that in male,and the expression of OBP4 in female was higher than that in female in ecolsion 0 h stage.It was speculated that OBP4 might be related to male courtship or refilled process of the heavy muscular area of the ductus ejaculatorius simplex,and OBP 17 might be related to allelopathy.(4)One hundred and twenty-three full-length genes were screened from olfactory related genes in GenBank.There are 14 complete odorant binding proteins(OBPs),4 complete sex pheromone binding proteins(PBPs),7 complete general odorant binding proteins(GOBPs),67 complete odorant receptors(ORs),4 complete chemosensory proteins(CSPs),1 complete gustatory receptor(GR),3 complete ionotropic receptors(IRs),2 complete odorant degrading enzymes(ODEs),6 complete sex pheromone degrading enzymes(PDE),2 protein-related genes,2 ejaculatory bulb-specific proteins,1 SNWA4 related protein and 1 NADPH oxidase related protein.The differential expression genes are are 8 OBPs,4 PBPs,5 GOBPs,23 ORs,3 CSPs,1 GR,2 IRs,2 ODEs,1 ejaculatory bulb-specific protein,1 SNWA4 related protein and 1 NADPH oxidase related protein.(5)At different developmental stages of C.sasakii,the most obvious expression chemosensory proteins were CSP18,OBP24,OBP30,OBP32,OBP35 and OBP40,and sex pheromone binding proteins were PBP2 and PBP3,and odorant binding proteins were OBP 12,OBP24,OBP29,OBP30,OBP31,OBP32,OBP35 and OBP40,and sex pheromone binding protein PBP2.The basic properties of chemosensory protein CSP22,odorant binding protein OBP40,and sex pheromone binding protein PBP3 were analyzed.The results showed that CSP22 was acidic and hydrophily,and the expression level was high in female and male adult after copulation.OBP40 is acidic protein and hydrophily.OBP40 protein predicts two ligands.The expression levels of Bom(hexadecyl-10-diene-1-ol)and Mg2+.OBP40 in males were higher than those in females.PBP3 is acidic and hydrophily.The expession of PBP3 in male is much higher than that in female.It is supposed to be related to the courtship of males.This study provides an important basic database for the study of the olfactory behavior of the C.sasakii,and lays a foundation for the further study of the behavior regulator of the C sasakii,and the scientific use of the behavioral regulation method to control the insect pests.
