| Common scab is one of the most important diseases in Chinese potato production.Traditional control measures cannot effectively prevent scabies and are not economical and environmentally friendly.Disease-resistant breeding is an economical and effective way to solve scab disease.Therefore,Streptomyce.Scabies(s.scabies),which is the most pathogenic pathogen of potato common scab,was used in this study to identify the resistance of 118 potato germplasm resources by pot inoculation.Then it has analyzed the correlation between tuber phenotypic traits and scab resistance.Through transcriptome sequencing technology,two copies of diploid potato tuber expression profiles of different resistance types were analyzed under the stress of scab,and differentially expressed genes were annotated and analyzed.Scab resistance genes were screened.Lay the theoretical foundation for molecular breeding of common scab.The main findings are as follows:1.Resistance screening obtained tetraploid material: 7 high resistant materials,68 resistant materials,30 susceptible materials,5 high susceptible materials and 7 heavy susceptible materials.Resistance screening obtained diploid material: 1 high resistant materials,2 resistant materials,1 susceptible materials,1 heavy susceptible material.2.The tuber phenotypic traits in 118 germplasm resources was statistically analyzed.There was a significant positive correlation between common scab index and lenticels density;There was a significant negative correlation between common scab index and tuber epidermal smoothness.There was a significant positive correlation between common scab index and skin color.There was a significant positive correlation between common scab index and fiesh color There was no significant correlation between resistance levels and tuber shape,eye depth.3.6515 and 6069 differentially expressed genes were identified for disease-resistant and susceptible varieties,respectively.There were more up-regulated differential genes and more resistant varieties than susceptible varieties.4.Differentially expressed genes in resistant varieties participate in major metabolic pathways: Plant-pathogen interaction and Carotenoid biosynthesis differentially expressed genes in susceptible varieties participate in major metabolic pathways: Plant-pathogen interaction and Carotenoid biosynthesis.Differential expression of carotenoid genes not only affects the skin color and fiesh color,but also affects the resistance level of common scab5.Transcription factors of WRKY,MYB,AP2/ERF transcription factor family are involved in the response to scab disease stress;10 candidate genes are screened in the plant-pathogen interaction pathway. |
