The Platycladus originated in China,is one of the most important tree species for afforestation on barren hills in China.This study of arborvitae champion tree seed quality were determined,arborvitae seed orchard to phenotypic arborvitae provenance index determination,determination of physiological and biochemical indexes,and by using SSR molecular marker using half a countryman family genetic diversity analysis,to our compatriots in different arborvitae half family related clustering analysis,in biota orientalis compatriots family save the evaluation and utilization to provide theoretical basis.The main findings are as follows:?1?Quality Evaluation of Platycladus orientalisAmong them,the highest 1000-seed weight was t-10#?22.91g?,the lowest 66#?13.63g?,and the average weight of each seed was 19.058g.The difference between the maximum and minimum value was 9.28g.The highest seed germination rate was 162#,the germination rate was 64.98%,the lowest was 49#,the germination rate was 2.91%,the average germination rate of each seed was 25.552%,the difference between the maximum and the minimum was39.428%.The highest seed germination potential was 162#,the germination potential was24.27%,the lowest was 170#,107#,49#,the germination potential was 0%,the average germination potential of each seed was 10.19%,and the difference between the maximum value and the minimum value was 24.27%.?2?Evaluation of half-sib family of Platycladus orientalis at seedling stageThe growth indexes of 15 half sibling families of cyperus rubii reached a significant level,among which the highest seedling height was 162#,the highest seedling height was45.86cm,the lowest seedling height was 66#,the lowest seedling height was 25.7cm,the average seedling height of each half sibling family was 36.71cm,and the difference between the maximum and minimum values was 20.16cm.The highest diameter was 167#,the ground diameter was 0.57cm,and the lowest was 66#,the ground diameter was 0.356cm.The index and seed weight,germination rate and germination potential.?3?Physiological and Biochemical Evaluation of Platycladus orientalisThe NR activity,soluble sugar content,chlorophyll content,and POD activity of Platycladus orientalis sibs were significantly different.The highest NR activity is 167#,with a content of 16.41?g·g-1h-1,and the lowest is 93#,with a content of 11.83?g·g-1h-1.The average NR activity of each Platycladus orientalis family is 14.20?g·g-1h-1,the difference between the maximum and minimum values is 4.58?g·g-1h-1;the highest soluble sugar content is 198#,the content is 956.25?g·g-1,the lowest is 66#,the content is 409.38?g·g-1,the average soluble sugar content of each Platycladus orientalis family is 631.81?g·g-1,the maximum and minimum values differ by 631.81?g·g-1;the highest chlorophyll content is 162#,the content is 7.08 mg·g-1,the lowest is 34#,the content is 1.94mg·g-1,the average chlorophyll content of each Platycladus orientalis family is relationship with growth and physiological and biochemical characteristics of semi-sibling families of Platycladus orientalisThere was a positive correlation between seed index and seedling growth index.In the determination of the relationship between physiological and biochemical indexes and growth indexes,the NR activity was positively correlated with the ground diameter.The soluble sugar content was positively correlated with seedling height and ground diameter.There was a significant positive correlation between chlorophyll content and seedling height,ground diameter and number of branches and leaves.POD activity was positively correlated with ground diameter.?5?Analysis of genetic diversity of Platycladus orientalis sibs based on SSR molecular markersThe average number of each of the 18 primer pairs was between 135 and 277,and the average amplified fragment size was 233.5556.The polymorphic bands of various sources were quite different.The genetic diversity of Platycladus orientalis provenances was significantly different,and the observed allele?Na?variation ranged from 3 to 17;The effective allele?Ne?ranged from 1.0076 to 2.7083;the Nei's gene diversity?H?ranged from0.0075 to 0.6308;The Shannon information index?I?ranged from 0.0249 to 1.3125.The 15Platycladus semi-sibling families averaged Na=6.1111,Ne=1.514,H=0.2761,I=0.5646.The number of alleles observed at the seed level of the half-sib family of Platycladus orientalis is Na=3.8331,the number of effective alleles Ne=0.5173,the diversity of Nei's genes H=0.2038,Shannon information index I=0.3868,Four parameters were significantly lower at the seed level than at the seed level.The degree of genetic differentiation was 0.1297,indicating that the degree of population differentiation was moderate.The average value of FIS statistic is-0.057,indicating the population heterozygous excess.The genetic differentiation coefficient calculated the gene flow level Nm=1.6775,indicating that there is moderate
