Wheat(Triticum aestivum)is the second largest food crop in China.Low temperature limits the distribution and production of crops worldwide.Me JA is a signaling molecule that plays an important role in response to abiotic stress in the plant.Dongongdongmai 1(Dn1),with a greening rate of 85%,is the first wheat variety that can safely overwinter in Heilongjiang Province.Dongnongdongmai 2(Dn2)is another wheat variety that is safe to overwinter in Heilongjiang Province after Dn1 and its cold resistance is stronger than Dongnongdongmai 1(Dn1).JM22 is a weak cold-resistant variety and cannot be wintered in heilongjiang province.In this study,the the strong cold-resistant winter wheat varieties Dn1,Dn2,and the weak cold-resistant wheat variety JM22 were used as experimental materials,and the Me JA treatment was applied externally at the three-leaf stage.The leaves and tillers were sampled when the temperature of the field dropped to 5?,0?,-10?,-25? under the treatment of Me JA.The effects of low temperature and Me JA on the relative expression of transcriptional repressors Ta JAZ7 and Ta JAZ12 were analyzed.Conduct a bioinformatics analysis of Ta JAZ7.The interaction relationship between Dn1 Ta JAZ7,Ta MYC2,and Ta ICE41 was preliminarily identified by yeast two-hetero method,providing theoretical basis and application support for the study of JAZ's participation in the cold resistance molecular mechanism of JA in winter wheat.The results showed:(1)Exogenous JA treatment significantly reduced the relative expression of Ta JAZ7 in three winter wheat tillers and leaves at-10?(except for Dn1 tillers),And significantly reduced the relative expression of Ta JAZ12 in tillers,and significantly increased the relative expression of Ta JAZ12 in leaves.(2)The CDS gene of Dn1 TaJAZ7 had a total length of 633 bp and encode 210 amino acids;the relative molecular weight of the protein is 21635.51 Daltons;it b elonged to the Tify family and has a Tify domain and a CCT-2 domain;the secondary structure of the protein was a mixed type;the protein was unstable,hydrophilic protein.(3)In vitro yeast two-hybrid results showed: Dn1 Ta JAZ7 and Ta MYC2,Ta ICE41 have physical interaction.
