Entomopathogenic fungi as a represent in the application of biological control is widely applied,Metarhizium robberstii ARSEF 23 as a model organism of entomopathogenic fungi is often used in the reseach of Insect-bacteria interaction,genetic regulation mechanism and so on.Histone deacetylase is closely related to fungal pathogenicity,sporulation regulation,genetic stability and resistance to adverse conditions,which contains a NAD'dependent histone deacetylase(SIRT2)gene in HDAC Family Class ?.In this study,we used the SIRT2 gene of Metarhizium robberstii as the research object and obtained the SIRT2 knockout strain by agrobacterium mediated homologous transformation mechanism.We proved that SIRT2 gene affects the stability of DNA and causes the degeneration of strain,but does not affect the virulence of Metarhizium robberstii,which provide experimental support.Meanwhile we analyze the function of SIRT2 gene in Metarhizium robberstii and provide a theoretical basis for the better use of Metarhizium robberstii in biological control in future.The results of the study are as follows:1.SIRT2 knockout plasmid and complement plasmid were successfully constructed,then SIRT2 gene knockout strain and SIRT2 supplementation strain were obtained.2.We have carried out a series of biological characteristics of three strains,to analyse the function of SIRT2 gene in Metarhizium robberstii.(1)In Metarhizium robberstii,the color patterns of SIRT2.knockout strains were all changed during the transfer of passages and were extremely unstable,which proved that SIRT2 gene affects the genetic stability of Metarhizium robberstii.(2)In Metarhizium robberstii,the SIRT2 gene is related to antioxidant capacity and the UV resistance of SIRT2 knockout strains have decreased significantly.(3)Compared with wild type Metarhizium robberstii,the infection ability of SIRT2 knockout strain to maize worm have not changed,meanwhile there was no difference in micromorphology of silkworm hemolymph.
