The developed olfactory system of insects is an important factor for the diversity of their populations,and olfactory genes play a central role in olfactory sensing,and have a direct impact on the foraging,mating,laying eggs and avoiding predators.Therefore,the study of insect olfactory molecular mechanism is of great theoretical significance for the development of insect olfactory based control technology.Apriona germari(Hope)is one of the important hardwood tree moth pests in China.It is destructive and causes serious economic loss and environmental damage.The strategy of insect olfactory management has been proposed and applied,but the olfactory system and its molecular mechanism of sensing have been rarely studied.Therefore,this paper provides a theoretical basis for the extraction and identification of the pheromone by observing the ultrastructures of the anterior thorax of A.germari.Further,this research focus on analysis of A.germari olfactory genes,and explore the A.germari olfactory receptor gene expression in different tissue between different sex,research results enrich the theoretical basis of chemical communication between A.germari,also in order to speed up the A.germari pheromone,or other volatile substances found provides a potential target genes and theoretical direction.The main results are as follows:(1)Suspected gland pores was found on the prothorax tissue of adult female A.germari by used VHX-6000 super deep scene 3D microscope and SEM.(2)Through bioinformatics analysis of female and male antennal transcriptome data of A.germari adults,62,299 Unigenes were obtained,with an average length of 948 bp.The successful functions of unigenes were annotated to Nr database,GO database,KO database and KOG database,with 36,834,27,260,16,846 and 16,044 respectively.There were 15,821 Unigenes with FPKM>10.(3)Using bioinformatics software to analyze and process antennae transcriptome data of A.germari,150 olfactory related genes were identified,including 62 odorant-binding proteins(OBPs),25 chemosensory proteins(CSPs),42 odorant receptors(ORs),3 ionotropic receptors(IRs),13 gustatory receptors(GRs)and 5 sensory neuron membrane proteins(SNMPs).(4)We examined the expression of the ORs and IRs genes in adult female and male antennae,maxillary palps,labial palps,and the end part of abdominal in A.germari by RT-q PCR.27 Ager OR showed an antenna-specific expression profile;15 Ager OR expressed in maxillary palps;25 Ager OR expressed in labial palps;38 Ager OR expressed in abdominal end,in particular,13 Ager OR showed high expression in male end body.Ager IR1 and Ager IR3 were found to express in the end body,Ager IR2 was highly express in the male labial palps.
