Japanese apricot(Prunus mume Sieb.et Zucc),which belongs to Rosaceae family.The ffuit is preferred due to nutritional importance and also used to prepare several value added products.Pistil is the basis for fruit formation.The present study was conducted to investigate the molecular mechanism.of pistil development in Japanese apricot cultivar,’Xiao lv e’.The process of pistil differentiation of’Xiao lv e’studied by paraffin section method.The fine structure of pistil observed by scanning electron microscope.Lignin content of flower without pistil(hollow flower or abortive pistil flower)and multi pistil flower(complete flower)of small green calyx was determined;CCoAOMT protein antibody prepared,and western blot reaction established for Japanese apricot protein.We collected the flower bud of’Xiao lv e’during growth stage.The RIP-seq test was carried out with CCoAOMT antibody.The main results are as follows:1.The early differentiation stage is the key period for multi pistil formation and primordia development.After the pistil differentiation completed,the abnormal development of pistils occurred.During flowering stage,the proportion ofXiao lv e’without pistil flowers(hollow flowers)was 18%.The relative content of lignin in flowers without pistils was significantly lower than that in flowers with pistils,indicating the lignin role in pistil development.2 We prepared the CCoAOMT protein antibody and established the Western-blot reaction system of plum.the results revealed that the polyclonal antibody of CCoAOMT protein was successfully prepared,and the direct extraction of SDS sample buffer solution checkedfor the extraction of protein from Japanese apricot.3.The RNA related to CCoAOMT protein was analyzed by RIP-seq Technology:Ctl(control group)and RIP(experimental group)obtained 57772404 and 46819880 original sequencing sequences(Raw reads)respectively.After filtration,57402032 and 46572600 sequences(Clean reads)were obtained.A total of 710 differentially expressed genes were screened5 outof which 203 were up-regulated and 507 down regulated.The genes associated with AP2 transcription factors(Prupe.6G126400)may be related to the development of flower organs.The gene associated with C3H transcription factors(Prupe.5G040400)may regulate the biosynthesis of lignin..GO enrichment analysis of these 710 differentially expressed genes showed that the total number of DEGs related to GO analysis were 476.The significant differentially expressed genes were mainly characterized in to three classes i.e.,biological processes and molecular functions.Seven genes were associated with glucan:glucosyltransferase activity.KEGG enrichment analysis depicted that biosynthesis of benzoxazine,interaction of plant and pathogen and the biosynthesis of secondary metabolites pathways found most enriched out of 63 pathways.The expression profiles indicated that the process of pistil development is the result of multi gene involvement.
