| In order to find out the effect of selenium deficiency on the physiological parameters and serum metabolism of Przewalski’s gazelles(Procapra przewalskii),to screen biomarkers with selenium deficiency diagnosis function,to explore the reasons why Procapra przewalskii did not appear Se-deficient symptoms in Se deficient areas,and to provide reference for the establishment of feeding standards for later artificial populations.In this experiment,the animal model of Se-deficient was established by feeding Se-deficient feed to Procapra przewalskii,included two processes,each of which has seven repetitions,collected the blood of jugular vein in different stages,the content of mineral elements,physiological and biochemical indexes in blood and metabolites in serum was detected.The results showed that(1)The Se content in the blood of the experimental group was extremely lower than that of the reference group(P<0.01),while the concentration of Cu was more than that of the control group(P<0.05),there was no significant difference in the content of Mn,Fe and Zn between the two groups(P>.05).(2)There were no significant differences in WBC,RBC,MCH,MCHC and Hb level between the se-deficient and control group(P>0.05).(3)The activities of GSH-PX in serum of P.przewalskii in experimental group was lower than that in reference group(P<0.05),but and T-SOD higher than that in reference group(P<0.05).CAT,T-AOC and MDA content were no obvious distinction between the two groups(P>0.05).(4)The PCA score of the metabolomics analysis showed that there was significant difference between Se-deficient and control group.The score of OPLS-DA showed that there was a significant dispersion and aggregation of metabolites between the two groups.The results of opls-da showed that there were 40 different metabolites and 53 different metabolites in the positive and negative ion mode.The main metabolic pathways involved in the biosynthesis of primary(secondary)bile acids and unsaturated fatty acids and aminoacyl tRNA and plant secondary metabolites,the metabolism of arginine and proline and pyrimidine,bile secretion,protein digestion and absorption,involved 26 different metabolites.We found that a single parameter,pseudouridine,was significantly associated(P=0.008)with P.przewalskii with Se-deficient,AUC of ROC analysis was 0.878,more than0.85,which is an effective diagnostic index.Comprehensive analyzed the blood physiological and biochemical indexes andmetabonomics results,the feeding of Se deficient feed had an impact on fat metabolism,energy metabolism,immune function and liver function of P.przewalskii,but there was a certain complementary function among various changed indexes,in addition,the fold-changes of different metabolites in serum were not high.Which are the reasons why P.przewalskii did not show the same Se deficiency symptoms as Tibetan sheep in the same basin. |
