Nicotiana tabacum is one of the important cash crops in the world,and it is a model plant that studies basic biological processes.Fructose-1,6-bisphosphate aldolase(FBA)is a key enzyme involved in plant glycolysis,gluconeogenesis,and the Calvin cycle.It plays an important role in biotic and abiotic stress responses and in regulating growth and development.Based on the N.tabacum genome information on the public genome platform,this study systematically identified and analyzed the characteristics of N.tabacum FBA genes.The expression of FBA in N.tabacum roots,stems and leaves,its response to light response,and its expression under abiotic stress were studied.The results of the study on the characteristics of N.tabacum FBA genes are as follows:(1)16 FBAs were identified in N.tabacum as class I FBAs,with conserved glycolytic domains,and subcellular localization revealed that NtFBA1-10 were localized to chloroplasts and NtFBA11-16 localized in the cytoplasm.Analysis of its physical and chemical properties,including protein ID,gene ID,protein length,protein molecular weight,isoelectric point of amino acid,average coefficient of hydrophilicity,and physical and chemical properties of NtFBAs genes are very similar.Only NtFBA6 and NtFBA10 are similar to Other members are different.(2)Phylogenetic analysis of NtFBAs and FBA genes of other species shows that NtFBAs were significantly different from Class II FBAs and had the highest homology with Class I FBA genes.Class I FBAs genes were divided into two branches,S1 and S2.NtFBA1-10 were located in S1,and NtFBA11-16 were located in S2.(3)Multiple sequence alignment of NtFBA genes and analysis of conserved motif.NtFBAs all had motif1/2/4/9,and these four motif belonged to the glycolytic domain.This high degree of conservation indicated that motif1/2/4/9 played an irreplaceable role in N.tabacum.(4)The intron-exon gene structure indicated that the number,length,and position of the intron-exons of the NtFBAs genes located in the same branch were very similar,but individual genes were also different.Most of NtFBAs in S1 were 4 to 6 introns,and only NtFBA6 contained 9 introns.Most of NtFBAs in S2 contained 2 introns,only NtFBA13 contained 17 introns,and NtFBA14 contained 16 introns.It was speculated that the phenomenon of intron acquisition in NtFBA6/13/14 genes occurred during evolution.(5)Studies on the expression levels of NtFBAs genes in root,stem and leaves and their response to light found that most NtFBAs genes were highest expressed in leaves.Most NtFBAs genes responded to light response,and the expression of FBA increased gradually with the increase of light time.The NtFBAs gene showed different expression patterns for 4 abiotic stresses,but it was consistent that the expression level of NtFBA13/14 were up-regulated,and the expression level of NtFBA7/8 were down-regulated in 4 abiotic stresses.
