Wheat leaf rust is a kind of wheat disease caused by Puccinia triticina(P.triticina)and is harmful to wheat production.P.triticina is a living nutritive fungi,and its specialization is very strong.In the incompatible combination,hypersentive reaction(HR)was considered to be the main defense response of wheat against P.triticina infection.Polyamines(PAs)is a class of biological metabolic molecules in organisms.Recent studies have found that PAs plays an important role in plant growth,development and responses to stress.A transcriptome database of the interaction between wheat and P.triticina was obtained.By analyzing the database,we found TaADC,a key gene in polyamine biosynthesis pathway,was expressed differently in incompatible and compatible combinations.On the basis of this,the wheat near-isogenic line TcLr26 and recurrent parent Thatcher(Tc)were used to generate incompatible and compatible combinations with P.triticina race 260.We analysis the function of TaADC in participating in defense response against P.triticina.The main results are as follows:1.The expression of TaADC was detect using qRT-PCR technology during the interaction between wheat and P.triticina.The results showed that TaADC was induced in the incompatible combinations,but did not change significantly in the compatible combinations.2.The subcellular localization vector carrying 35S::TaADC-GFP recombinant plasmid was constructed and transiently transformed into tobacco.It was found that TaADC was localized in the cytoplasm.3.The TaADC gene was silenced in wheat with VIGS and RNAi technology,respectively,then the P.triticina race 260 was inoculated.Compared with the control,the host HR area of the single invasion point of the plant after silencing was increased,and the number of HMC of P.triticina was also increased.This indicates that the host resistance is weakened after silencing the TaADC gene.
