ABC transporters,namely ATP-binding cassette transporter?ABC?,is an ancient family of transporters.Most ABC transporters are major active transporters.They can bind and hydrolyze primary active transporters of ATP,and transport a large number of substrates across lipid membranes.Two subtypes of ABC transporters,ABCB and ABCC,are responsible for transporting a variety of molecules,including peptides,iron,bile salts,drugs,toxic substances and phospholipids.ABCB and ABBC are widely distributed in prokaryotes and eukaryotes.They have conservative domains and perform a variety of biological functions.In this study,the full length of ABCB and ABCC genes were cloned and identified in mussel,and their expression under the stress of lipopolysaccharide?LPS?,copper(Cu2+)and benzopyrene(B[a]p was analyzed.The aim of this project is to provide theoretical basis for further understanding the stress mechanism of mussel transport proteins under external stress,and also provide molecular targets for future stress resistance breeding.Specific research results are as follows:Mc-ABCB and Mc-ABCC have high homology with other species ABCB and ABC,including Walker A motif,Walker B motif,Q-loop,D-loop,H-motif,the ABC-signature motif?LSGGQ-motif?and other functional domains.ABCB gene and ABC gene were expressed in seven tissues of mussel,and the highest expression was found in digestive gland and parotid tissue.Under the stimulation of LPS,the expressions of ABCB gene and ABCC gene in the gills of mussels increased gradually,and returned to normal level with the passage of stimulation time.Under Cu2+ion stimulation,the expression of Mc-ABCBmRNA gradually increased with time,showing an adaptive trend;the expression of Mc-ABCC in parotid region increased significantly at first,and decreased slowly with the time of stimulation.The expressions of Mc-ABCB and Mc-ABCC increased in different degrees under the stimulation of benzopyrene.The results showed that both Mc-ABCB and Mc-ABCC were involved in the stress process of mussel response to external stimuli,and could be used as molecular targets for stress resistance breeding of mussel.The results also provide a theoretical basis for the possibility of Mc-ABCB and Mc-ABCC as biomarkers of environmental pollution.
