Trichinella spiralis can cause immune response in BALB/c mice during the early phase of infection,and start the immune regulation mechanism.However,most studies stays in the middle or late stage of infection,and the phenomenon and underlying mechanism of the early stage of infection is still not thorough enough.In order to investigate how Trichinella infection induces intestinal pathological changes and the expression of cytokines related to immune regulation in the early stage.In this study,the identification of male and female muscle larva under a microscope was used to purify and count the worms,and reproductive capacity index of Trichinella spiralis was observed with different doses of ML attack,the pathological changes of intestinal tract in early stage of Trichinella spiralis infection in BALB/c mice were observed by hematoxylin and eosin staining(HE),immune cells and cytokines profiles of T.spiralis infected mice were examined by Meso Scale Discovery(MSD)and flow cytometry.The results of the RCI showed that the fecundity of 250 random ML was stable,determining it as the most suitable infection dose for attacking insects.The HE results of intestine showed obvious intestinal mucosa thickening and eosinophils infiltration.The MSD results of the spleen showed that the expression of IL-2 and IFN-?was significantly increased at 3 days post-infection(dpi),and decreased at 6 dpi;the expression of IL-4 and IL-10 increased significantly at 3 dpi to 6 dpi.The mesenteric lymph node showed the expression of IL-4 increased significantly 6 dpi.Flow cytometry analysis showed that the level of CD4~+/CD8~+at 30 h after infection was lower than that at 6 h after infection(P<0.05),the level of IL-4 was markedly decreased at 30 h after infection(P<0.01),IL-17 was significantly increased at 6 dpi(P<0.05),the levels of IFN-?and Foxp3~+were significantly increased 6 dpi(P<0.01).The results showed that:the pathological changes of intestinal tract of BALB/c mice appeared at 6 h after infection,and there was a hybrid immune response with significant expression of Th2 type immune cell marker cytokines at 3 d dpi.Trichinella spiralis plays an immune regulatory role by inducing the high expression of Th2 type immune cell marker cytokines in the host.
