Cloning of the gene PsnHB13 and PsnHB15 of the HD-ZIP family,the cDNA of PsnHB13 is 870bp encoding 289 amino acids.The cDNA of PsnHB15 is 2559bp encoding 852 amino acids.Both are the transcriptome differential genes for the PsnCycD1;1 transgenic poplar(Populu ssimonii × P.nigra).With the increasing of the PsnCycD1;1 expression,it also increases for PsnHB13 and PsnHB15 refer to the results of analysis of differences in expression.Tissue specificity analysis shows that the highest gene expression of them is in the poplar stem.The result of yeast two-hybrid tecnique suggests,PsnHB13 interacts with both of PsnHB15 and PsnCycD1;1.But there is no interaction beteween PsnHB15 and PsnCycD1;1.After getting the construction of overexpression vectors(pROKII-PsnHB13 and pROKII-PsnHB15),Plant overexpression vectors were constructed and transformed into wild-type tobacco by agrobacterium-mediated leaf disc transformation.Compared with wild-type tobacco,we can find that smaller and longer leaves,the slow growth of roots and smaller flowers in PsnHB13 transgenic tabaccos.And It indicates that PsnHB15 plays a negative role in the growth of tobacco leaves,roots and flowers.Meanwhile,it was found in PsnHB15 transgenic tabaccos of uneven and curly blades.So PsnHB15 has an effect on growth and development of tobaccco leaf to change shape of tobacco leaves.By poplar transformation for PsnHB13 and PsnHB15,the ratio between leaf lengh and leaf width increases in early leaf.However,the difference will disappear with growth of tansgenic poplar.It indicates that both of the PsnHB13 and PsnHB15 have effect on growth and development of poaplar blades to change shape of poplar leaves.What is more,the ratio between dry weight and fresh weight of leaves and stems increases to some degree in PsnHB13 transgenic poplar which gets shorter for wild-type one.And wen can infer that PsnHB13 plays a positive role on dry matter accumulation of leaves and stems and a negtive role on growth and development of poplar stems.By construction of vectors of pRNAi-PsnHB13 and pRNAi-PsnHB15 and poplar genitic transformation,compared with wild-type poplar,PsnHB13 expression drops down to 1.16%furthest.And PsnHB15 expression drops down to 1.11%furthest.
