Seed germination is an early stage of plant growth and development,which is an important and critical period.This process is relatively complicated and involves a series of complex physiological and metabolic processes.It is one of the most important issues to study mechanisms of seed germination.Research object of the test was the bloated seed of Cupressus gigantea,compared and analyzed the germination rate under different treatments.We analyzed the proteomics of the seed germination.The results showed that:1)the germination rate were the highest after the fresh seed soaked in the 20? water,cultured in the incubator with 16 hours of light which temperature is 20?.Seeds were soaked by 1.Omg/mL GA3 had the greatest effect on the germination.2)It was divided into four stages according to the phenotype of the seed:the seed swelling is the first stage;the seed coat is broken is the second stage;the radicle break through the endosperm is the third stage;the radicle elongate to the bottom become red and the top become green is the fourth stage.We found 34 proteins were expressed differentially,13 proteins found in the third stage and 17 proteins found in the fourth stage.The biological processes include:cytoskeleton dynamics(10,29.41%),energy metabolism(5,14.71%),transcription and translati-on(5,14.71%),stress response(5,14.71%),molecular chaperones(3?8.82%),Amino acid metabolism(3?8.82%),oxidation reduction(2,5.88%),and participation in the ABA signaling pathway(1,2.94%).These different function proteins were interacted to provide essential nutrients for seed development.Among them,storage proteins play an extremely important role.Through the studies,we optimized the germination conditions of Cupressus gigantea seeds.Established the theoretical basis for the study of proteomics during germination.
