Microorganisms play an important role in forest.In this study,bacterial flora analysis of pear canker,poplar bacterial canker and willow wilt disease spot tissue was carried out.Four suspected new taxa of bacteria were isolated and obtained.Polyphasic taxonomy study was carried out on these four strains of bacteria to determine their taxonomic status and laid a foundation for the development and utilization of these bacteria in the future.The research results are as follows:The composition of bacteria in the diseased tissue of pear canker is complex.In this study,more than ten kinds of bacteria were isolated from the diseased tissue samples,belonging to 9different genera.Based on the analysis of the bacterial flora of the diseased tissue of poplar bacterial canker,nearly 100 strains of bacteria were isolated at the junction of disease and health,belonging to 36 different genera and 58 bacterial species.Among these bacteria isolated,the four genera of Pseudomonas,Microbacterium,Enterobacter and Bacillus have the most bacterial species.During the isolation and identification of bacterial population in the diseased tissue of willow bacterial wilt disease,the dominant population Brenneria salicis was isolated from the diseased tissue.Through inoculation experiments,it was found that Brenneria salicis isolated from poplar and willow had different pathogenicity.?1?Description of Herbaspirillum piri sp.nov.sh Q-4Tis aerobic,motile bacterial strain.Cells are 0.8?m×1.5?m in size.The colonies are pale yellow,smooth.After testing strains tolerance in different growth condition,the strain can not grow when condition temperature is bellow 10°C and above 41°C.Growth occurs from p H4.0 to 8.0 and between 1 and 3%?w/v?Na Cl.The major fatty acids of this strain were C16:0,C16:1?6c/C16:1?7c,cyclo-C17:0and C18:0.The respiratory quinone of sh Q-4Tcontents ubiquinone 8?94%?and the Q-7?6%?.The DNA G+C content is 60.4mol%.Thus,strain sh Q-4Trepresents a novel species of the genus Herbaspirillum.The type strain is sh Q-4T?=CFCC 14641T=LMG 52804T?,isolated from the bark of a pear tree.?2?Description of Sinorhodobacter populi sp.nov.sk2b1Tis Gram-staining negative,non-motile,facultative anaerobic.The cell is1.3?m-1.5?m.Their growth occurred between 10 and 41°C and at p H 5.0 to 7.0.Additionally,growth occurred in conditions of 0-5%?w/v?salinity,but not above 7%Na Cl.The major fatty acids are C16:0?8.8%?and C18:1?7c/C18:1?6c?79.43%?.The main respiratory quinone was Q-10.The type strain is sk2b1T?=CFCC 14580T=KCTC 52802T?.?3?Description of Ancrocorticia populi sp.nov.Sk1b4Tis a Gram-staining positive facultative anaerobic,non-motile strain.The cells is a0.4?m-0.6?m long and 0.3?m-0.4?m wide short rod bacteria.Growth of the novel strain occurred at temperatures between 10°C and 41°C,and within a p H range of 6.0-9.0;optimal growth occurred at 30°C.Growth also occurred within a Na Cl concentration of 0%to 5%?w/v?.The major fatty acids of the strain were C14:0?41.3%?,C16:0?27.2%??C18:1?9c?9.9%?.The type strain is sk1b4T?=CFCC 14564T=KCTC 39919T?.?4?Description of two subspecies of Brenneria salicisGrowth of CFCC 11710Toccurred at temperatures between 10°C and 41°C,and within a p H range of 5.0-9.0;optimal growth occurred at 30°C.Growth also occurred within a Na Cl concentration of 0 to 3%?w/v?.The major fatty acids of the strain are C16:1?6c/C16:1?7c?22.52%?,C16:0?24.74%?and C18:1?7c/C18:1?6c?22.84%?.The DNA G+C content is 53mol%.So the strain was classified new subspecies and named it Brenneria salicis subsp.populi subsp.nov.The type strain is CFCC 11710T.Growth of CFCC 14753Toccurred at temperatures between 10°C and 37°C,and within a p H range of 6.0-9.0;optimal growth occurred at 30°C.Growth also occurred within a Na Cl concentration of 0 to 3%?w/v?.The major fatty acids of the strain are C16:1?6c/C16:1?7c?23.13%?,C16:0?24.22%?and C18:1?7c/C18:1?6c?20.83%?.C18:0ante/C18:2?6,9c?0.63%?is notable fatty acid of this subspecies.The DNA G+C content is 53mol%.Named it Brenneria salicis subsp.salicis subsp.nov.The type strain is CFCC 14753T=LMG 6144T.
