Abstract/Summary:
Ericerus pela is a kind of scale insect of Hemiptera,Coccoidea,which is an important resource insect in China.Symbiotic bacterias play an important role in host insects,such as providing nutrients to host insects,regulating host insect growth and reproduction,and increasing the resistance of host insects.In this paper,high-throughput sequencing technology was used to sequence the V3-V4 region of 16 S r DNA of symbiotic bacteria in E.pela.In this study,high-throughput sequencing technology was used to sequence and annotate the V3-V4 region of 16 S r DNA of symbiotic bacteria in various developmental stages of E.pela.The study mainly got the following results:(1)Species identification and classification of symbiotic bacteria in E.pela were completed:There are various types of symbiotic bacteria in the E.pela.624 OTUs were found in 10 groups of samples.There were 22 phylum,36 classes,75 orders,125 families,210 genera,and95 species were annotated.(2)Analysis of dominant and sub-dominant bacteria in various developmental stages of E.pela:The dominant bacteria in the developmental stage of the E.pela is Rickettsia,with an average relative content of 94.88%.Among them,the male adult of E.pela have the highest content,and second-instar females have the lowest.Also high in content are Methylobacterium,Sphingomonas,and the like.Compared with other insects,the dominant bacteria of E.pela is Rickettsia,and the content is extremely high.However,Rickettsia has not been found as a dominant strain in other studied insect.Secondly,symbiotic bacteria commonly found in other insects such as Buchnera and Wolbachia have not been detected in white E.pela.(3)Analysis of the dynamic changes of symbiotic bacteria in E.pela:The species and abundance of the sub-dominant bacteria in different developmental stages of E.pela are quite different,but the content of Methylobacterium and Sphingomonas was always higher during the development of E.pela.And there are many sub-dominant bacteria unique to some developmental stages in the male worms.The α-diversity of E.pela was the highest in the second-instar female stage,and the female adult was the lowest in the early stage.Among the males,the male eggs were the highest and the male adults were the lowest.Through β-diversity analysis,it was found that the species difference of the symbiotic bacteria community in the developmental stage of the male was higher than that of the female,and the species distribution of the symbiotic bacteria was closely related to the sex of the E.pela insect.In summary,this study explored the diversity of symbiotic bacteria in the developmental stages of E.pela,the changes of community during development,and the differences of symbiotic bacteria between male and female in the same developmental stage.We conducted a preliminary discussion on the factors that contribute to the differences in the community of symbiotic bacteria.This laid the foundation for further research on how E.pela and their symbiotic bacteria interact with each other.
